HPLC法测定小鼠血清中的4-甲基-3-硝基苯甲酸及药动学研究

【摘要】:目的:建立4-甲基-3-硝基苯甲酸(4-methyl-3-nitro-benzoic acid,MNBA)的HPLC测定方法,进行药动学研究。方法:用乙腈直接沉淀蛋白提取样品;采用Intertex C18色谱柱(150 mm×4.6 mm,5μm),以磷酸溶液(p H 0.3)-乙腈(50∶50)为流动相,流速1.0 m L·min-1,检测波长230 nm;25℃条件下对流动相、血清样品预处理方法进行筛选,并对该方法进行方法学验证。结果:待测物质分离度良好,质量浓度在测定范围内与峰面积线性关系良好(r2≥0.999 9),方法精密(日内、日间精密度均≤5%)且准确(低、中、高3种质量浓度的准确度均在95%~110%之间),低、中、高3种质量浓度的样品在4℃冰箱保存10 d,-20℃保存20 d及反复冻融3次后均保持稳定无降解。小鼠腹腔注射MNBA后在体内的代谢过程符合开放一房室模型(w=1/c),其药代动力学参数为:T1/2分别为(17.0±3.2)h、(30.5±2.7)h和(80.7±2.0)h;AUC(0-t)分别为(7 801.8±2 360.7)mg·L-1·min-1、(16 589.1±5 383.6)mg·L-1·min-1和(56 898.9±12 153.5)mg·L-1·min-1。结论:经方法学验证,本法可作为检测MNBA的测定方法;本文测定的腹腔注射MNBA在小鼠体内的药动学过程为MNBA的后续研究提供参考。
