angiopep-2修饰的脑靶向制剂研究进展

【摘要】：脑部疾病化学药物治疗的关键是穿透血脑屏障(BBB)。受体介导的胞吞转运是目前递送药物入脑应用广泛且较成熟的策略。在BBB和脑胶质瘤细胞上低密度脂蛋白受体相关蛋白-1(LRP-1)高表达,angiopep-2(ANG)可与LRP-1特异性结合实现脑靶向性,对脑肿瘤具有双级靶向效应,且靶向效率高于乳铁蛋白和转铁蛋白等靶向分子。本文介绍了ANG的结构与功能,及其在脑靶向制剂中的应用,为脑靶向制剂的研发提供依据,为脑部疾病的治疗提供参考。
[Abstract]:The key to chemotherapeutic therapy for brain diseases is to penetrate the blood-brain barrier (BBB).) Receptor-mediated endocytosis is a widely used and mature strategy for drug delivery into the brain. Low density lipoprotein receptor associated protein-1 (LRP-1) is highly expressed in BBB and glioma cells. Angiopep-2 (ANG) can specifically bind to LRP-1 to achieve brain targeting, and has a two-stage targeting effect on brain tumors. The targeting efficiency was higher than that of lactoferrin and transferrin. In this paper, the structure and function of ANG and its application in brain targeting preparation are introduced, which provide the basis for the research and development of brain targeting preparation and provide reference for the treatment of brain diseases.
