氨茶碱诱导斑马鱼幼鱼产生运动失调

【摘要】:氨茶碱因促进内源性肾上腺素与去甲肾上腺素释放而对呼吸道平滑肌有直接松弛作用而用于哮喘病的治疗,但其不良反应和过敏反应常会引起患者死亡。为了考察氨茶碱的对斑马鱼幼鱼运动失调的影响,本研究使用不同浓度氨茶碱溶液于24孔板中直接作用于72 hpf孵化的斑马鱼幼鱼,定时间记录各组幼鱼的抽搐频率、心率变化、体形变化等参数,用于评价斑马鱼幼鱼运动情况。结果表明,氨茶碱诱导的斑马鱼幼鱼开始时运动增加,随后伴有自发转动,随着药物作用时间延长,出现典型的抽搐反应而产生严重的颤抖,斑马鱼幼鱼出现运动失调轻重程度和快慢呈时间和浓度依赖性。从本实验结果推测,氨茶碱应慎用或不得用于婴幼儿和小孩哮喘病的治疗,也要慎用于孕期妇女的用药治疗。
[Abstract]:Aminophylline is used in the treatment of asthma because of its direct relaxation effect on respiratory smooth muscle due to promoting the release of endogenous epinephrine and norepinephrine, but its adverse reactions and allergic reactions often cause death in patients. In order to investigate the effect of aminophylline on the movement disorders of juvenile zebrafish, different concentrations of aminophylline solution were directly applied to juvenile zebrafish hatched for 72 hpf in a 24-well plate. The frequency of convulsion and heart rate were recorded at a fixed time. Body shape change and other parameters were used to evaluate the movement of juvenile zebrafish. The results showed that the motility of juvenile zebrafish induced by aminophylline increased at the beginning, followed by spontaneous rotation. Juvenile zebrafish movement imbalance degree and speed are time and concentration dependent. The results suggest that aminophylline should be used with caution or not in the treatment of infant and child asthma, but also in pregnant women.
