BAPTA-AM脂质体抗大鼠重症急性胰腺炎的作用

【摘要】:为观察BAPTA-AM脂质体(BA-L)抗重症急性胰腺炎(SAP)的作用,采用胰胆管逆行注射4%牛黄胆酸钠(TC)制作大鼠SAP模型.BA-L治疗组于造模后立即尾静脉注射相应剂量BA-L,给药24h后,观察各组大鼠的死亡情况,检测各组大鼠血清AMS,LDH,TNF-α水平以及胰腺组织匀浆MDA水平及SOD活性,HE染色观察胰腺组织病理学改变.结果表明:给予BA-L治疗后SAP大鼠死亡率明显降低,血清AMS,LDH,TNF-α水平和胰腺组织匀浆MDA水平显著降低,SOD活性增高,HE染色组织切片显示BA-L明显减轻胰腺组织损伤.表明BA-L能显著减轻大鼠SAP,为开发BA-L成临床SAP治疗新药物提供必要的药效学依据.
[Abstract]:To observe the effect of BAPTA-AM liposome (BA-L) on (SAP) in severe acute pancreatitis. The rat model of SAP was established by retrograde injection of 4% sodium bovine cholate (TC) into the pancreatic duct. The death rate of rats in the BA-L treatment group was observed 24 hours after the injection of the corresponding dose of BA-L, into the tail vein immediately after the establishment of the model. The levels of serum AMS,LDH,TNF- 伪, MDA level and SOD activity of pancreatic homogenate were measured in each group. The histopathological changes of pancreas were observed by HE staining. The results showed that the mortality of SAP rats was significantly decreased, the serum AMS,LDH,TNF- 伪 level and pancreatic homogenate MDA level were significantly decreased, and the activity of SOD was increased after BA-L treatment. HE staining showed that BA-L significantly alleviated pancreatic tissue injury. The results showed that BA-L could significantly reduce rat SAP, and provide the necessary pharmacodynamic basis for the development of BA-L as a new drug for clinical SAP treatment.
