RP-HPLC法测定去甲斑蝥素齐墩果酸复合脂质体的包封率

【摘要】:目的:建立测定去甲斑蝥素齐墩果酸复合脂质体包封率的方法。方法:采用反相高效液相色谱法。色谱柱为Thermo Sycronis C18,流动相为磷酸水溶液(p H=3.2)-甲醇(梯度洗脱),流速为1.0~1.7 ml/min,检测波长为210 nm,柱温为30℃,进样量为50μl。结果:去甲斑蝥素、齐墩果酸的质量浓度分别在20~300、10~150μg/ml范围内与峰面积呈良好线性关系(r=0.999 8、0.999 7);精密度、稳定性、重复性试验的RSD≤5.89%;去甲斑蝥素的平均回收率为99.15%~100.11%,RSD≤3.20%(n=5),齐墩果酸的平均回收率为99.22%~100.30%,RSD≤3.34%(n=5);去甲斑蝥素、齐墩果酸平均包封率分别为(48.01±2.34)%、(87.31±0.56)%(n=5)。结论:该方法法简便、快速、可靠,可用于去甲斑蝥素齐墩果酸复合脂质体包封率的测定。
