氢氧化钙缓释肠溶胶囊的给药动力学及其解酒抗醉评价

【摘要】:为了了解氢氧化钙缓释肠溶胶囊的给药特性及应用,本文采用体外模拟法考察了氢氧化钙缓释肠溶胶囊的给药动力学,并对其解酒抗醉作用进行评价。结果表明,氢氧化钙的代谢受其释放速度控制,负载量在3.2~5.5 mmol/g范围,其溶解度稳定在10 mg/100m L左右,释放半衰期在13~45 min,并随负载量的增大而缩短,缓释为分层释放,各层符合零级释放模型;家兔急毒试验表明氢氧化钙缓释肠溶胶囊安全无毒,解酒抗醉结果为解酒组和抗醉组的醉睡时间分别为163 min和125 min,比模型组的259 min明显缩短,氢氧化钙缓释肠溶胶囊具有较强的解酒抗醉能力。
[Abstract]:In order to understand the characteristics and application of calcium hydroxide sustained-release enteric-soluble capsules, the pharmacokinetics of calcium hydroxide sustained-release enteric-coated capsules was investigated by in vitro simulation method, and the antiintoxication effect of calcium hydroxide sustained release enteric capsules was evaluated. The results showed that the metabolism of calcium hydroxide was controlled by the release rate of calcium hydroxide, the loading amount was in the range of 3.2g / 5.5 mmol/g, the solubility of calcium hydroxide was about 10 mg/100m / L, and the half life of calcium hydroxide was 1345 min, and shortened with the increase of loading amount. The slow release is layered release, each layer accords with zero order release model. The acute toxicity test of rabbits showed that calcium hydroxide sustained-release enteric-coated capsule was safe and non-toxic. The antiintoxication time of antiintoxication group and anti-intoxication group were 163 min and 125 min, respectively, which were significantly shorter than that of the model group (259 min). Calcium hydroxide sustained-release enteric capsule has strong antiintoxication ability.
