24孔板培养活跃链霉菌合成那西肽条件的优化

【摘要】:目的优化24孔板培养活跃链霉菌XW产那西肽的发酵条件。方法采用单因素实验和响应面法对24孔板的发酵条件进行优化。结果得到活跃链霉菌XW 24孔板发酵产那西肽的最优发酵条件为:种子培养基装液量2.55 m L,发酵培养基装液量0.93m L,发酵时间121h。结论通过对发酵条件的优化,可以明显提高24孔板发酵产那西肽的效价达到882.25μg/m L,实验值与模型预测值较接近,吻合度较高,较初始效价675.63μg/m L提高了30.58%。研究结果表明24孔板发酵与摇瓶发酵之间有良好的相关性,相关系数R为0.883。
[Abstract]:Objective to optimize the fermentation conditions of natoxin produced by Streptomyces XW in 24-well plate culture. Methods single factor experiment and response surface method were used to optimize the fermentation conditions of 24-well plate. Results the optimum fermentation conditions were as follows: the amount of seed culture medium was 2.55 mL, the amount of fermentation medium was 0.93m L, and the fermentation time was 121 h. The optimal fermentation conditions of Streptomyces actives XW 24-well plate fermentation were as follows: the amount of liquid in seed culture medium was 2.55 mL, the fermentation time was 121 h. Conclusion through the optimization of fermentation conditions, the titer of nacitide produced by 24-well plate fermentation reached 882.25 渭 g / mL, the experimental value was close to the predicted value of the model, and the coincidence degree was higher. Compared with the initial titer of 675.63 渭 g / mL, it was increased by 30.58%. The results showed that there was a good correlation between 24-well plate fermentation and shaking flask fermentation, and the correlation coefficient was 0.883.
