固相萃取-HPLC法测定人血清中10-羟基卡马西平的浓度

【摘要】:目的:建立测定人血清中10-羟基卡马西平(MHD)浓度的方法,促进临床个体化用药和相关药动学研究。方法:血清样品采用固相萃取,用高效液相色谱法进样测定。色谱柱为Venusil C18,流动相为甲醇-水(45∶55),流速为0.8 ml/min,紫外检测波长为230 nm。结果:本试验中内源性物质不干扰测定,MHD血药浓度在2.49~39.82 mg/L范围内线性关系良好(r=0.999 8),检测限为0.4 mg/L;高、中、低(2.49、19.91、39.82 mg/L)质量浓度平均提取回收率为97.2%~101.2%,平均方法回收率为98.3%~100.8%,日内和日间RSD均10%。结论:该方法快速、稳定、准确,适用于MHD的血药浓度测定和相关药动学研究。
