基于PLGA-PEG载体材料的铂类药物纳米输送

【摘要】:铂类药物为临床最广泛应用的一类抗肿瘤药物,但使用过程中出现的肾毒性、耳毒性和耐药性等,给铂类药物的长期使用带来了挑战。采用纳米载体靶向输送铂类药物是提高药物疗效、降低不良反应的有效方式。聚乳酸-羟基乙酸共聚物(PLGA)-聚乙二醇(PEG)嵌段共聚物是纳米载体普遍使用的高分子材料之一,具有生物相容性好、可生物降解、可组装成纳米粒等优点,日益受到关注。以PLGA-PEG嵌段共聚物为载体材料,输送二价铂配合物或四价铂配合物,可达到被动或主动靶向肿瘤部位、提高疗效并减轻不良反应的目的。此外,简单讨论了PLGA-PEG材料在铂类药物输送中的发展方向及存在的问题。
[Abstract]:The platinum-based drugs are the most widely used anti-tumor drugs, but the nephrotoxicity, ototoxicity and drug resistance in the use process have brought the challenge to the long-term use of the platinum-based drugs. The use of the nano-carrier to target the platinum-based medicine is an effective way to improve the curative effect of the medicine and reduce the adverse reaction. Polylactic acid-glycolic acid copolymer (PLGA)-polyethylene glycol (PEG) block copolymer is one of the widely used polymer materials of the nano-carrier, and has the advantages of good biocompatibility, biodegradability, and can be assembled into nanoparticles. The PLGA-PEG block copolymer is used as a carrier material to deliver a bivalent platinum complex or a tetravalent platinum complex to achieve the purpose of passive or active targeting of the tumor site, improving the curative effect and reducing the adverse reaction. In addition, the development direction and the existing problems of PLGA-PEG materials in the delivery of platinum drugs are discussed briefly.
