蒽醌类衍生物C3通过ERK1/2-ERCC1信号通路对结肠癌细胞影响的研究

细胞外调节蛋白激酶(extracellular regulated protein kinases,ERK1/2)级联是一种经典的细胞内信号通路,对多种细胞生命活动的调控及癌症的发生至关重要。在级联反应中,ERK1/2磷酸化会激活各种转录因子,调控细胞生理学过程。切除修复交叉互补组1(excision repair cross-complementation group 1,ERCC1)是ERK1/2下游转录因子之一,识别损伤DNA并修复。研究发现,用蒽醌类衍生物大黄素靶向作用ERK1/2抑制其下游ERCC1的表达,可以抑制癌症恶化。蒽醌类衍生物是以9,10-蒽醌为骨架的一种天然酚类,具有良好的抗癌活性。目前已有多种蒽醌类衍生物作为抗癌药物应用于临床,且取得了不错的效果。但是,蒽醌类衍生物水溶性较差,在自然界中含量有限且提取程序复杂。因此,通过化学方法设计合成新型的蒽醌类衍生物,为抗癌药物的研发提供了新的思路。本课题组前期以2-甲基蒽醌为原料,经过一系列化学反应合成了一种易溶于水的蒽醌类衍生物(1-硝基-2-酰基蒽醌-缬氨酸,简称C3),并发现其对结肠癌细胞具有抑制作用,但具体分子机制...
