交通相关PM2.5对人群外周血细胞因子的影响及钙信号通路调控研究

【摘要】:目的: 探讨太原市交通相关PM2.5对人群外周血细胞因子的影响,并初步探讨交通相关PM2.5通过外周血淋巴细胞Ca2+-CaN/IRAK-1—NFAT信号通路调控细胞因子,对深入探讨PM2.5的免疫毒性机制提供参考依据。 方法: 选择太原市外勤交通警察为目标人群,疾病预防控制中心(CDC)工作人员为对照人群,监测两人群工作环境中PM2.5浓度,采集并分离两人群外周血淋巴细胞,采用荧光分光光度法检测不同人群淋巴细胞内[Ca2+]i的变化;ELISA法检测人群血清中细胞因子TNF-α、IL-1、IL-2、IL-4的表达水平;荧光定量PCR(Q-PCR)法测定细胞钙调磷酸酶(CaN)、白介素-1受体相关激酶-1(IRAK-1)、活化T细胞核因子(NFAT)、IL-2和TNF-α基因的mRNA表达;Western Blotting测定CaN、IRAK-1、NFAT蛋白表达情况。 结果: 太原市交警工作环境中PM2.5浓度(0.132mg/m3)高于CDC人员工作环境中PM2.5浓度(0.100mg/m3)(P0.05)。交警淋巴细胞内钙离子浓度高于CDC人员。交警淋巴细胞内IL-2mRNA表达比CDC工作人员低(P0.05);男性交警TNF-α mRNA表达水平比CDC工作人员高(P0.05)。交警血清中细胞因子TNF-α、 IL-1表达水平比CDC工作人员高(P0.05);而交警血清IL-2表达水平比CDC工作人员低(P0.05);交警淋巴细胞内CaN mRNA和IRAK-1mRNA表达水平比CDC工作人员有增高趋势,交警淋巴细胞内NFAT mRNA表达水平比CDC工作人员高(P0.05)。血清浓度与淋巴细胞内Ca2+浓度、血清TNF-α浓度与CaN和NFAT基因表达均有正相关关系(P0.05),血清IL-2水平与Ca2+呈负相关关系(P0.05)。 结论: 太原市交通相关PM2.5可增加外周血IL-1和TNF-α细胞因子表达,抑制IL-2分泌,外周血淋巴细胞Ca2+-CaN/IRAK-1-NFAT信号通路可能参与TNF-α的调控。
[Abstract]:Objective:. To investigate the effect of traffic related PM2.5 on peripheral blood cytokines in Taiyuan, and to explore the regulation of cytokines by traffic related PM2.5 through the Ca2 -CaN / IRAK-1-NFAT signaling pathway in peripheral blood lymphocytes, which provides a reference for the further study of the immunotoxicity mechanism of PM2.5. Methods:. The field traffic police in Taiyuan city were selected as the target population and the staff of the CDC as the control group. The concentration of PM2.5 in the working environment of the two populations was monitored and the peripheral blood lymphocytes were collected and separated from the two populations. The changes of [Ca2] I in lymphocytes of different populations were detected by fluorescence spectrophotometry and the expression level of cytokine TNF- 伪, IL-1and IL-2IL-4 in serum of different populations was detected by enzyme-linked immunosorbent assay (Elisa). The expression of calcium phosphatase (Cam), interleukin-1 receptor associated kinase (IL-1) and IRAK-1was detected by fluorescence quantitative PCR- Q-PCR method. The mRNA expression of IL-2 and TNF- 伪 gene was activated by T cell nuclear factor, and the expression of CaNNRAK-1nFAT protein was detected by Western Blotting. Results:. The concentration of PM2.5 in the working environment of traffic police in Taiyuan City was 0.132 mg / m ~ (3) higher than that in the working environment of CDC personnel (0.100 mg / m ~ (3)) and the concentration of Ca ~ (2 +) in the lymphocytes of traffic police was higher than that of CDC personnel. The expression of IL-2mRNA in the lymphocytes of traffic police was lower than that of CDC workers (P 0.05); the concentration of IL-2mRNA in the lymphocytes of male traffic police was lower than that of CDC workers. The expression level of mRNA was higher than that of CDC staff. The expression level of cytokine TNF- 伪 and IL-1 was higher than that of CDC staff, while the level of IL-2 in traffic police was lower than that of CDC staff. The expression level of CaN mRNA and IRAK-1mRNA in the lymphocytes of traffic police was higher than that of CDC staff. There is a tendency for CDC staff to increase, The expression level of NFAT mRNA in the lymphocytes of traffic police was higher than that of the CDC workers. There was a positive correlation between the serum concentration of TNF- 伪 and the expression of CaN and NFAT gene, and a negative correlation between the level of serum IL-2 and Ca2. Conclusion:. Traffic related PM2.5 in Taiyuan can increase the expression of IL-1 and TNF- 伪 cytokines and inhibit the secretion of IL-2. The Ca2 -CaN / IRAK-1-NFAT signaling pathway in peripheral blood lymphocytes may be involved in the regulation of TNF- 伪.
