饲喂不同浓度D-果糖对大鼠生理机能的影响

【摘要】:在过去的20年间,果糖作为甜味剂在食品工业中的使用量从20%增加至30%。随着产量与用量迅速增长,人们在逐渐认识到果糖独特优点的同时,也开始关注果糖的营养特性与食用安全性。近年来果糖相关的营养学问题、食用安全性问题以及人体内代谢途径等方面的研究成为学界研究的热点。本文用含10%、20%、30%果糖的饲料喂养28日龄的大鼠,以含10%葡萄糖的饲料和基础饲料饲喂大鼠作参比,探讨不同浓度的果糖饮食对大鼠机体健康的影响,得到以下结论: 1、每4d观察记录大鼠采食量、体重的变化,发现不同浓度的果糖和低浓度的葡萄糖饮食,未导致大鼠体重上升和肥胖,且果糖浓度与大鼠体重呈负相关关系,高浓度果糖饮食导致大鼠体重偏轻。 2、每14d检测大鼠血清低密度脂蛋白(LDL)、高密度脂蛋白(HDL)、胆固醇(CHO)、甘油三酯(TG)、瘦素(LP)、胰岛素(INS)的含量,发现在大鼠未成年阶段,果糖和葡萄糖对大鼠血清LDL,HDL, CHO,TG含量的影响较大。不同浓度的果糖,对大鼠的血液指标的影响有所不同。当低浓度果糖进入机体后,使血清LDL、HDL、CHO及TG含量都升高,HDL和LDL的升高幅度没有明显差异。而高浓度果糖饮食刺激LDL和HDL含量上升同时,HDL/LDL下降,且比低浓度果糖更大幅地使CHO含量上升,但对TG含量没有明显的影响。高低浓度的果糖饮食都能引发LP抵抗和INS升高,且低浓度果糖更明显。 3、喂养至第56d时,采集大鼠的肝脏样本,称重并检测其中碱性磷酸酶(ALP)、硬脂酰辅酶A去饱和酶(SCD)、乙酰辅酶A羟化酶(ACC)、丙酮酸激酶(PK)和脂肪酸合成酶(FAS)的含量,发现高低浓度的果糖饮食都促进肝脏脂质的合成,且高浓度的果糖摄入导致肝脏病变。 试验发现不同浓度的果糖饮食能导致大鼠不同生理指标的变化,影响生长发育。
[Abstract]:Over the past 20 years, the use of fructose as a sweetener in the food industry has increased from 20% to 30. Attention has also been paid to the nutritional characteristics and safety of fructose. In recent years, there have been nutritional problems related to fructose, The study on food safety and metabolic pathway in human body has become a hot topic in academic circles. In this paper, the rats of 28 days of age were fed with a diet containing 10 ~ 20% fructose and fed with a diet containing 10% glucose and a basic diet as a reference. To study the effects of fructose diet with different concentrations on the body health of rats, the following conclusions were obtained:. 1. The changes of intake and body weight of rats were observed every 4 days. It was found that different concentrations of fructose and low concentration of glucose diet did not cause weight gain and obesity in rats, and there was a negative correlation between fructose concentration and body weight of rats. High fructose diet resulted in low body weight in rats. 2. The contents of LDLX, HDLN, CHOX, TGN, LPX, INS were detected every 14 days in rats, and it was found that the contents of LDLX, HDLX, CHOX, TGN, LPX, INS) in rats were found to be in infancy. Fructose and glucose had a great effect on the contents of HDL and CHOG in serum of rats. Different concentrations of fructose had different effects on blood indexes of rats. When low concentrations of fructose entered the body, There was no significant difference between LDL and HDL-CHO and TG, but high fructose diet stimulated the increase of LDL and HDL content and decreased HDL / HDL content, and increased CHO content more than that of low fructose concentration. However, there was no obvious effect on TG content. Fructose diet with high or low concentration could induce LP resistance and INS increase, and low fructose concentration was more obvious. 3. At the 56th day of feeding, the liver samples of rats were collected, and the contents of alkaline phosphatase (ALPN), stearyl coenzyme A desaturase (SCD), acetyl coenzyme A hydroxylase (ACCK), pyruvate kinase (PKK) and fatty acid synthase (FASs) were measured. It was found that fructose diet promoted liver lipid synthesis, and high fructose intake led to liver lesion. It was found that different concentrations of fructose diet could lead to the changes of different physiological indexes and affect the growth and development of rats.
