同煤职工外周血白细胞染色体相对端粒长度与高血压相关性研究

【摘要】:目的:本研究旨在了解煤矿工人外周血端粒长度与原发性高血压之间的关系,为煤炭工人预防高血压提供科学依据。方法:1.使用QIAGEN公司TIA Namp Blood mini kit(51104)试剂盒提取测试对象外周血白细胞基因组DNA;2.使用实时荧光定量PCR测定端粒长度,T/S值为相对端粒长度,即端粒长度与单拷贝基因比值;3.运用SPSS19.0软件对受试样本进行数据分析,研究其外周血端粒长度与高血压的相关性。结果:1.本研究为配对设计实验,高血压病例组和对照组各230例。高血压组平均收缩压为145.73±16.99mm HG,平均舒张压为91.87±14.37mm HG,对照组平均收缩压为120.07±10.23mm HG,平均舒张压为76.24±8.71mm HG,两组存在显著差异。2.高血压病例组和对照组在年龄、身高方面无显著差异;而在总胆固醇(TC)、谷氨酰转肽酶(GGT)、谷氨酸(GLU)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)、甘油三酯(TG)、总蛋白(TP)、BMI方面,差别具有统计学意义,高血压病例组各指标均高于对照组。3.高血压病例组外周血端粒长度变化范围是0.2149-1.9531,对照组外周血端粒长度的变化范围为0.4562-2.1673,两组相比高血压组端粒长度明显缩短(P0.001)。外周血白细胞端粒长度与年龄呈负相关(P0.05)。女性高血压患者的端粒平均长度为1.2613±0.4112,男性高血压患者端粒平均长度为0.9981±0.3965。男女高血压患者端粒平均长度上存在显著差异(P0.05)。结论:1.高血压病例组在总胆固醇(TC)、谷氨酰转肽酶(GGT)、谷氨酸(GLU)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)、甘油三酯(TG)、总蛋白(TP)、BMI方面的指标上高于对照组,差别具有统计学意义(P0.05)。2.端粒长度与年龄呈负相关,年龄越大,端粒长度越短。男性高血压端粒平均长度低于女性高血压端粒平均长度。3.高血压人群端粒长度与正常人群的端粒长度有差异,高血压人群的相对端粒长度较正常人群短(P0.05)。
[Abstract]:Objective: to investigate the relationship between peripheral blood telomere length and essential hypertension in coal miners, and to provide scientific basis for preventing hypertension in coal miners. Method 1: 1. TIA Namp Blood mini kit (51104) of QIAGEN Company was used to extract genomic DNA from peripheral blood leukocytes. The T / S value of telomere length measured by real-time fluorescence quantitative PCR was relative telomere length, that is, the ratio of telomere length to single copy gene was 3. The correlation between peripheral blood telomere length and hypertension was studied by SPSS19.0 software. The result is 1: 1. This study was a pair design experiment, the hypertension case group and the control group 230 cases each. The mean systolic blood pressure (SBP) and diastolic blood pressure (DBP) were 145.73 卤16.99mm and 91.87 卤14.37mm in hypertension group, 120.07 卤10.23mm and 76.24 卤8.71mm in control group, respectively. There was significant difference between the two groups. There was no significant difference in age and height between the hypertension group and the control group, but there was no significant difference between the two groups in terms of total cholesterol, glutamyl transpeptidase (GGTN), glutamate GLU, high density lipoprotein (HDL), low density lipoprotein (LDLN), triglyceride (TGN), and total protein (TTP) BMI. The difference was statistically significant. The indexes of hypertension group were higher than that of control group. 3. 3. The range of telomere length in patients with hypertension was 0.2149-1.9531, and that in control group was 0.4562-2.1673.The telomere length in the two groups was significantly shorter than that in the hypertensive group (P 0.001). The telomere length of peripheral blood leukocytes was negatively correlated with age (P 0.05). The mean telomere length was 1.2613 卤0.4112 in female hypertensive patients and 0.9981 卤0.3965 in male hypertensive patients. There was significant difference in mean telomere length between male and female patients with hypertension (P 0.05). Conclusion 1. The indexes of TC, GGT, GLU, HDLX, LDLX, TGG, TGN, TG, TG, TG and TG in hypertension group were higher than those in control group, and the difference was statistically significant (P0.05. 2) in the hypertension group, there was no significant difference between the two groups in the index of total cholesterol, glutamyl transpeptidase, high density lipoprotein (HDL), low density lipoprotein (LDLN), triglyceride (TGN) and total protein (TGP). Telomere length was negatively correlated with age, and the older the telomere length was, the shorter the telomere length was. The mean telomere length of male hypertension is lower than that of female hypertension. The telomere length of hypertensive population was significantly different from that of normal population, and the relative telomere length of hypertensive group was shorter than that of normal population (P0.05).
