绿原酸对断奶应激大鼠免疫功能和肠道屏障功能的影响

【摘要】:本实验旨在探讨绿原酸对断奶大鼠免疫功能和肠道屏障功能的影响。选用32只健康的21±1日龄断奶SD大鼠(62.26±2.73g),按体重随机分成4个组:正常组,LPS组(第13天1mg/Kg.BW LPS),CHA20组(每天20mg/kg CHA),CHA50组(每天50mg/kg CHA),每组8只大鼠,试验期为14天。本实验首先研究绿原酸对断奶大鼠不同时间段内生长性能的影响,随后对断奶大鼠在13天注射脂多糖加强应激,研究绿原酸干预对断奶大鼠脂多糖应激生理功能、免疫功能以及肠道屏障功能的影响。 实验结果表明,CHA50组对断奶大鼠生长性能有较好的的促进作用,能显著降低7-14d料肉比(p0.05);脂多糖应激断奶大鼠后导致小肠长度指数、脾重指数、血清IgM水平、空肠和回肠绒毛高度和绒腺比、结肠和空肠Occludin和ZO-1蛋白表达、结肠和空肠组织IL-10水平显著降低(p0.05)以及显著增加(p0.05)血清IL-2和IFN-γ水平、IFN-γ/IL-4比值、血清尿素氮和白蛋白浓度、血清DAO浓度、空肠和回肠隐窝深度、尿液中L/M比值、结肠和空肠组织中IFN-γ和TNF-α的含量。绿原酸能显著增加(p0.05)小肠长度指数、脾重指数、血清IgM含量、空肠和回肠绒毛高度和绒腺比、结肠和空肠Occludin和ZO-1蛋白表达、空肠组织IL-10水平;显著降低(p0.05)血清IL-2和IFN-γ水平、IFN-γ/IL-4比值、尿素氮和谷丙转氨酶浓度、空肠和回肠隐窝深度、尿液中L/M比值、血清DAO浓度、结肠和空肠组织中IFN-γ和TNF-α的含量。 综上所述,绿原酸能促进断奶大鼠的生长发育,缓解断奶大鼠造成的免疫力降低、肠道形态结构和屏障功能损伤,降低肠道通透性,调节肠道的炎症反应,提高机体的免疫功能以及肠道屏障功能。
[Abstract]:The aim of this experiment was to investigate the effects of chlorogenic acid on the immune function and intestinal barrier function of weaned rats. 32 healthy SD rats (62.26 + 2.73g) were randomly divided into 4 groups according to weight: normal group, LPS group (thirteenth days 1mg/Kg.BW LPS), CHA20 group (20mg/ kg CHA every day), CHA50 group (50mg/kg CHA), 8 rats in each group. The period was 14 days. The effect of chlorogenic acid on the growth performance of rats in different periods of weaning was first studied. Then the rats were injected with lipopolysaccharide for 13 days to strengthen stress, and the effects of chlorogenic acid on the physiological function, immune function and intestinal barrier function of LPS in weaned rats were studied.
The experimental results showed that the CHA50 group had a better promoting effect on the growth performance of the weaned rats and significantly reduced the ratio of 7-14d to meat (P0.05). After the lipopolysaccharide stressed weaning rats, the length index of the small intestine, the spleen weight index, the serum IgM level, the height of the jejunum and ileum villus and the ratio of the cashmere gland, the expression of Occludin and ZO-1 in the colon and jejunum, and the colon and the colon were observed. IL-10 levels in the jejunum tissue were significantly reduced (P0.05) and significantly increased (P0.05) serum IL-2 and IFN- gamma levels, IFN- gamma /IL-4 ratio, serum urea nitrogen and albumin concentration, serum DAO concentration, jejunum and ileum depth, urine L/M ratio, the content of IFN- gamma and TNF- alpha in the colon and jejunum tissue. Chlorogenic acid could significantly increase the length of the small intestine (P0.05) small intestine length Degree index, spleen weight index, serum IgM content, jejunum and ileum villus height and cashmere gland ratio, Occludin and ZO-1 protein expression in colon and jejunum, IL-10 level in jejunum, serum IL-2 and IFN- gamma level, IFN- gamma /IL-4 ratio, urea nitrogen and glutamic transutase concentration, jejunum and ileum depth, L/M ratio in urine, L/M ratio in urine, D serum, serum D. AO concentration, IFN- and TNF- alpha in colon and jejunal tissues.
To sum up, chlorogenic acid can promote the growth and development of weaned rats, alleviate the decrease of immunity caused by weaning rats, the damage of intestinal morphology and barrier function, reduce the intestinal permeability, regulate the intestinal inflammatory reaction, improve the immune function of the body and the intestinal barrier function.
