灵芝孢子粉对亚慢性染镉大鼠肝脏损伤的保护作用

【摘要】:目的:研究灵芝孢子粉对亚慢性染镉大鼠肝脏损伤的保护作用,并探讨其可能的保护途径。方法:健康雄性SD大鼠84只,体重(90±10)g,随机分为:正常对照组(C)、单独染镉组(Cadmium,Cd)和2个给药剂量组(灵芝孢子粉0.5g/kg,1.0g/kg+Cd Cl2,即GCd L和GCdH组),每组各21只。大鼠适应性喂养7d后,每天对GCdL组和GCdH组大鼠灌相应剂量的灵芝孢子粉,C组和Cd组灌等体积蒸馏水。Cd组、GCdL组和GCd H组每隔一天腹腔注射一次氯化镉(1.0mg/kg),每周称重1次,并观察大鼠的一般情况,并在第30天、60天、90天分批处死,每组各处死7只;颈动脉取血,快速取肝脏,生理盐水冲洗,滤纸吸干多余的水分,称重,记录,-80℃保存;全自动血生化仪检测大鼠丙氨酸氨基转移酶(Alanine aminotransferase,ALT)和天门冬氨酸氨基转移酶(Aspartate transaminase,AST)的浓度;光学显微镜观察大鼠肝脏病理组织学变化;比色法检测肝脏丙二醛(Malondialdehyde,MDA)含量;酶联免疫吸附试验(ELISA)检测大鼠肝脏IL-6含量;Real time RT-PCR检测大鼠肝脏p65 m RNA、Mt-1 m RNA、Mt-2 m RNA的表达量。结果:与正常对照组比较,60d、90d的Cd组大鼠体重明显降低(P0.05),给予灵芝孢子粉干预后,大鼠体重均有不同程度增加,在90天时,GCdH组大鼠体重明显高于Cd组(P0.05);与空白组相比,大鼠染镉后肝脏脏器系数明显增大(P0.05),给药剂量组大鼠肝脏脏器系数与Cd组比较均有不同程度降低(P0.05);与空白组相比,单独染镉组ALT、AST明显升高(P0.05),而灵芝孢子粉各剂量组ALT、AST均有不同程度降低,且与单独染镉组比较差异有统计学意义(P0.05);HE染色结果显示,单独染镉组肝组织细胞间质充血严重,细胞排列杂乱,细胞界限不清晰,而给药组以上情况均有不同程度改善,并且高剂量组改善情况优于中剂量组;比色法检测MDA结果显示,经镉处理大鼠肝脏MDA含量较空白组明显增高(P0.05),给予灵芝孢子粉干预后,在60d、90d均有降低(P0.05);在30、60、90天,与空白组相比,单独染镉组大鼠肝脏细胞因子IL-6(白介素6)含量明显升高(P0.05);随着时间的延长,Cd组、GCdL组、GCdH组IL-6含量也逐渐增加(P0.05);与单独染镉组相比,给药组大鼠肝脏Mt-1 m RNA、Mt-2 m RNA的表达量明显升高(P0.05),而p65 m RNA表达明显降低(P0.05)。结论:灵芝孢子粉对亚慢性染镉大鼠肝脏损伤有保护作用,其机制可能与抗氧化应激,降低炎性细胞因子IL-6表达和诱导MT产生有关。
[Abstract]:Aim: to study the protective effect of ganoderma lucidum spores powder on liver injury in rats with subchronic cadmium exposure and explore its possible protective pathway.
