抗放利质量控制方法及辐射防护机制的研究

【摘要】:随着核能和辐射技术在能源、工农业生产、医疗卫生、科学研究和国防建设等领域中的应用日益广泛,许多国家对辐射损伤的预防和救治都极为重视,对辐射损伤防护药物的合成、筛选以及作用机制的研究也日益兴盛起来。 抗放利,化学名为2,2-二甲基四氢噻唑盐酸盐,是我所研制的具有自主知识产权的一类新药,对急性辐射损伤有较好的防护作用。目前国内外缺少此化合物的质量控制方法,对其辐射防护作用机制的研究较少,本研究采用专属性较强的气相色谱法、高效液相色谱法进行了鉴别试验,采用高效液相色谱法建立了抗放利含量测定的方法,气相色谱法建立了有机溶剂残留的检查方法,AccQ-Tag柱前衍生的方法建立了有关物质的检查方法,并按照药典的要求进行了方法学验证。 本研究选择了辐射敏感的造血系统和免疫系统作为研究对象,通过外周血象、MDA含量、SOD活性等指标评价了抗放利对正常小鼠的辐射防护作用,通过小鼠的生存时间、外周血指标、肿瘤生长曲线等指标评价抗放利对荷瘤小鼠的辐射防护作用。实验结果提示,抗放利可明显保护照射引起的免疫器官和造血功能的损伤,使造血功能恢复显著加快,增强机体的抗氧化能力,减轻脂质过氧化反应;抗放利能够有效地延长受照荷瘤小鼠的生存时间,使造血功能恢复明显加快,但不降低肿瘤组织的辐射敏感性。 实验结果表明,已建立的研究方法专属性强、准确性高、精密度好,可以作为抗放利质量控制的方法,为进一步制定抗放利质量控制标准提供了必要的基础和依据;抗放利具有较好的辐射防护作用,能够选择性地保护正常组织,其作用机制可能与其保护造血组织、提高机体免疫功能和抗氧化作用有关,该论文的相关研究为深入研究抗放利的作用机制奠定了基础。
[Abstract]:With the increasing application of nuclear energy and radiation technology in the fields of energy, industrial and agricultural production, medical and health care, scientific research and national defense construction, many countries attach great importance to the prevention and treatment of radiation damage. Studies on the synthesis, screening and mechanism of radiation protection drugs have become increasingly popular. The chemical name is 2o 2 dimethyl tetrahydrothiazole hydrochloride. It is a kind of new drug with independent intellectual property rights developed by me. It has good protective effect on acute radiation injury. At present, there is a lack of quality control methods for this compound at home and abroad, and the mechanism of radiation protection is less studied. In this study, the specific gas chromatography and high performance liquid chromatography (HPLC) were used to identify the compound. A method for the determination of radioimmunoassay was established by HPLC, and a method for the determination of residual organic solvents was established by gas chromatography. The method for the determination of related substances was established by pre-column derivation of AccQ-Tag. The methodology was validated according to the requirements of pharmacopoeia. In this study, radiation sensitive hematopoietic system and immune system were selected as the research objects. The radiation protective effect of anti-radiation on normal mice was evaluated by the indexes of MDA content and SOD activity in peripheral blood. The survival time and peripheral blood index of mice were also studied. Tumor growth curve was used to evaluate the radiation-protective effect of anti-radiotherapy on tumor-bearing mice. The results suggest that anti-radiation can obviously protect the damage of immune organs and hematopoietic function induced by irradiation, accelerate the recovery of hematopoietic function, enhance the antioxidant ability of the body, and alleviate the lipid peroxidation. Anti-radiotherapy can effectively prolong the survival time of irradiated mice and accelerate the recovery of hematopoietic function, but it does not reduce the radiosensitivity of tumor tissue. The experimental results show that the established research method has strong specificity, high accuracy and good precision. It can be used as a method for quality control of anti-reaping interest, and provides the necessary basis and basis for the further development of quality control standard of anti-reaping interest. The mechanism of anti-radiation protection may be related to the protection of hematopoietic tissue, the enhancement of immune function and the anti-oxidation effect, and the mechanism may be related to the protection of hematopoietic tissue, the enhancement of immune function and the anti-oxidation effect of the body. The related research in this paper lays a foundation for further study on the mechanism of anti-release effect.
