补体片段C3f对人胚肺成纤维细胞合成α-平滑肌肌动蛋白的影响

【摘要】:目的观察补体片段C3f对人胚肺成纤维细胞(MRC-5)合成α-平滑肌肌动蛋白(α-SMA)的影响。方法将低中高3个不同质量浓度(15.6,62.5,250 ng·mL-1)人工合成的补体片段C3f(实验组)加入培养基中,以刺激MRC-5细胞,以细胞免疫组化法检测α-SMA在MRC-5细胞质中的表达情况。同时,设立对照组:10 ng·mL~(-1)转化生长因子-β_1(TGF-β_1)和空白组(浓度为0的C3f),进行对比研究。结果中剂量实验组与对照组(TGF-β_1)刺激MRC-5细胞合成α-SMA量分别为7.46±0.66,7.54±0.51,差异无统计学意义(P0.05),说明2组的量相当,表明C3f具有刺激MRC-5合成α-SMA的功能。低中高3个不同质量浓度的细胞内α-SMA表达量分别为4.95±0.78,7.46±0.66和9.23±0.21,显示C3f的浓度与α-SMA的表达量呈正相关(P0.05),表明C3f具有促进MRC-5表达α-SMA的功能。结论补体片段C3f能够刺激MRC-5细胞内α-SMA的合成增加。
[Abstract]:Objective to observe the effect of complement fragment C 3f on the synthesis of 伪 -smooth muscle actin (伪-SMA) in human embryonic lung fibroblasts (MRC-5). Methods three synthetic complement fragments C3f (experimental group) of low, medium and high concentration (15.6n 62.5250 ng mL-1) were added to the medium to stimulate MRC-5 cells, and the expression of 伪 SMA in the cytoplasm of MRC-5 was detected by immunohistochemical method. At the same time, 10 ng mL~ (-1) transforming growth factor- 尾 _ 1 (TGF- 尾 _ 1) and control group (C _ 3f with concentration of 0) were set up for comparative study. Results the amount of 伪-SMA stimulated by TGF- 尾 _ 1 in the middle dose group and the control group was 7.46 卤0.666 卤7.54 卤0.51, respectively. There was no significant difference between the two groups (P0.05), indicating that C3f had the function of stimulating MRC-5 to synthesize 伪 -SMA. The expression levels of 伪-SMA were 4.95 卤0.78 卤7.46 卤0.66 and 9.23 卤0.21, respectively, indicating that C3f was positively correlated with the expression of 伪-SMA (P0.05), indicating that C3f could promote the expression of 伪-SMA by MRC-5. Conclusion C3f can stimulate the synthesis of 伪-SMA in MRC-5 cells.
