纳米氧化石墨烯颗粒对小鼠心肌酶的影响

【摘要】:目的探讨一次性尾静脉注射纳米氧化石墨烯颗粒(GO)对小鼠心肌ATP酶、LDH、AST活性影响,为其在生物学领域的安全应用提供科学依据。方法选择80只清洁级ICR小鼠,体重(18±2)g,雌雄各半,随机分为4组,分别以剂量为1.40、0.70、0.35 mg/kg氧化石墨烯对小鼠一次性尾静脉注射,对照组注射蒸馏水。并于注射后3 d和15 d将小鼠脱臼处死,取心脏,测定心肌匀浆液中的Na~+,K~+-ATP酶、Ca~(2+),Mg~(2+)-ATP酶、LDH、AST活性测定。结果注射后3 d,雌、雄中、高剂量Na~+,K~+-ATP酶活性及Ca~(2+),Mg~(2+)-ATP酶活性均高于对照组(P0.05)。15 d后,雌、雄各剂量组Na~+,K~+-ATP酶及Ca~(2+),Mg~(2+)-ATP酶活性均低于对照组(P0.05)。注射后3 d,雌、雄各组间AST活性无统计学意义(P0.05),15 d后,雌、雄各剂量GO组明显低于对照组(P0.05)。注射3 d,雄性各剂量GO组LDH活性明显低于对照组(P0.05),雌性高剂量GO组明显低于对照组和低剂量GO组。注射后15 d,雄性高剂量GO组显著低于中、低剂量GO组(P0.05),雌性中、高剂GO量组明显低于对照、低剂量GO组(P0.05)。结论一次性尾静脉注射氧化石墨烯可干扰Na~+,K~+-ATP酶、Ca~(2+),Mg~(2+)-ATP酶和AST、LDH等心肌酶活性的作用。
[Abstract]:Objective to investigate the effect of single tail vein injection of nano-graphene oxide granule (GO) on the activity of ATP and LDH,AST in myocardium of mice, and to provide scientific basis for its safe application in biological field. Methods Eighty clean grade ICR mice, weighing (18 卤2) g, half male and half female, were randomly divided into 4 groups. The mice were injected with graphene oxide at a dose of 1.40U 0.700.35 mg/kg via a single tail vein, while the control group was injected with distilled water. The mice were killed 3 and 15 days after injection, and the Na~, K- ATP, Ca~ (2), Mg~ (2)-ATP and LDH,AST activities in myocardial homogenate were determined. Results the activities of Na~, K- ATP, Ca~ (2), Mg~ (2)-ATP in female and male were higher than those in control group 3 days after injection (P0.05). 15 days after injection, Na~, K- ATP enzyme and Ca~ (2) in female and male groups were higher than those in control group. The activity of Mg~ (2)-ATP was lower than that of control group (P0.05). There was no significant difference in AST activity between female and male groups at 3 d after injection (P0.05). 15 days later, the activity of GO in female and male groups was significantly lower than that in control group (P0.05). The LDH activity of male GO group was significantly lower than that of control group (P0.05), and that of female high dose GO group was significantly lower than that of control group and low dose GO group. 15 days after injection, male high dose GO group was significantly lower than middle dose group, low dose GO group (P0.05), female, high dose GO group was significantly lower than control group, low dose GO group (P0.05). Conclusion single tail vein injection of graphene oxide can interfere with the activities of myocardial enzymes such as Na~, K- ATP, Ca~ (2), Mg~ (2)-ATP and AST,LDH.
