水解乳清蛋白对辐射损伤大鼠蛋白质营养状态及抗氧化能力的影响

【摘要】:目的 乳清蛋白属于优质蛋白,是临床肠内营养配方食品的优质蛋白质来源。水解乳清蛋白是乳清蛋白经酶促反应水解后的产物,所含的小分子短肽和游离氨基酸可在肠道不经消化而被直接吸收,较乳清蛋白有更多生物活性。本文通过就水解乳清蛋白对辐射损伤大鼠蛋白质营养状态及抗氧化能力影响的研究,旨在为其临床应用提供依据。 方法 清洁级SD大鼠40只(雌雄各半),鼠龄14周,体重269.7±15.2g,随机分为正常对照组、实验对照组、实验Ⅰ组、实验Ⅱ组,每组10只。正常对照组、实验对照组饲料氮源为乳清蛋白,实验Ⅰ组、实验Ⅱ组饲料氮源为水解乳清蛋白,实验Ⅱ组大鼠同时每天灌胃10%富硒SOD绿茶溶液[茶粉1g/(kg·d)],各组饲料其他组成均参照AIN-76、AIN-93配方配制。实验对照组、实验Ⅰ组和实验Ⅱ组动物给予一次性6Gray剂量的60Co-γ射线全身辐照。记录每天大鼠的进食量,,定期监测体重,21天后收集72小时粪便,采集血液并处死动物,测定粪氮,计算蛋白质AD值,进行血清TP、ALB和MDA含量及T-AOC、SOD、GSH-PX活力等相关指标测定,并进行肠黏膜HE染色。 结果 1、正常对照组动物整个实验期摄食量正常,体重持续增加。实验对照组、实验Ⅰ组、实验Ⅱ组动物辐照后摄食量和体重均明显低于正常对照组(p<0.01),实验Ⅰ组、实验Ⅱ组动物体重分别在辐照9天后、6天后开始增加,并且实验Ⅱ组动物体重非常显著高于实验对照组(p<0.01),也显著高于实验Ⅰ组(p<0.05)。 2、实验对照组、实验Ⅰ组、实验Ⅱ组粪氮排出量均非常显著高于正常对照组(p<0.01),但实验Ⅰ组、实验Ⅱ组非常显著低于实验对照组(p<0.01)。在实验动物辐射损伤情况下AD值计算结果表明,实验对照组、实验Ⅰ组、实验Ⅱ非常显著低于正常对照组(p<0.01),而实验Ⅰ组、实验Ⅱ组非常显著高于实验对照组(p<0.01)。 3、实验对照组血清TP和ALB含量均显著低于正常对照组(p<0.01)。实验Ⅰ组、实验Ⅱ组血清TP、ALB含量虽然低于正常对照组,但仅血清ALB含量在实验Ⅰ组与正常对照组间存在统计学差异(p<0.05),并且实验Ⅰ组、实验Ⅱ组血清ALB含量非常显著高于实验对照组(p<0.01)。 4、实验对照组血清T-AOC、T-SOD、GSH-PX活力均非常显著低于正常对照组(p<0.01)。实验Ⅰ组、实验Ⅱ组血清T-AOC和GSH-PX活力也非常显著低于正常对照组(p<0.01),但非常显著高于实验对照组(p<0.01),并且实验Ⅱ组血清GSH-PX活力也非常显著高于实验Ⅰ组(p<0.01)。实验对照组、实验Ⅰ组、实验Ⅱ组血清MDA含量均非常显著高于正常对照组(p<0.01),但实验Ⅰ组、实验Ⅱ组非常显著低于实验对照组(p<0.01)。 5、实验各组肠黏膜HE染色也显示不同程度的病理学改变。 结论 1、6Gray剂量的60Co-γ射线一次性全身辐照可严重影响受试动物营养状态。 2、水解乳清蛋白的消化、吸收与利用明显优于乳清蛋白,更有利于改善辐射损伤大鼠的蛋白质营养状态。 3、水解乳清蛋白可以减轻辐照大鼠的过氧化损伤,与富硒SOD绿茶粉联合应用效果更好。
