黑大豆种皮提取物对酒精性肝损伤的干预作用

【摘要】:黑大豆不仅具有较高的营养价值,且含有多种植物化学物。花色苷是黑大豆的主要生物活性物质。花色苷具有抗氧化、抗炎、改善视力、改善脂代谢、保护心血管等诸多保健功效。目前尚缺少黑大豆种皮提取物对酒精所致化学性肝损伤干预作用的报道。 目的: 以小鼠急性酒精性肝损伤为模型,通过对小鼠肝脏还原性谷胱甘肽、肝脏丙二醛、甘油三酯含量测定、肝脏组织病理切片等分析,探讨黑大豆种皮提取物对酒精性肝损伤的影响。 材料与方法: 雄性昆明小鼠,随机分为正常对照组、酒精模型组、黑大豆种皮提取物低剂量组(30mg·kg-1)、黑大豆种皮提取物高剂量组(60mg·kg-1)。按照《保健食品检验与评价技术规范(2003版)》的要求,每日灌胃一次,第31天,除正常对照组外,一次性灌胃给予50%无水乙醇溶液,造成急性酒精性肝损伤,禁食不禁水12h后检测各组小鼠的肝脏系数、肝脏甘油三酯(TG)、肝脏丙二醛(MDA)、脑组织MDA、肝脏还原型谷胱甘肽(GSH)、全血GSH的含量,观察肝脏的病理组织学改变并进行肝组织脂肪病变评分。应用SPSS18.0统计软件,进行单因素方差分析或秩和检验。 结果: 1.黑大豆种皮提取物高剂量组动物的肝脏组织GSH含量,明显高于酒精肝损伤模型组(P<0.05),且与正常对照差异不明显。黑大豆皮提取物低剂量组动物的肝脏组织GSH含量与肝损伤模型对照组比较,差异不明显。 2.黑大豆皮提取物低剂量组和高剂量组动物的肝脏组织MDA含量均低于酒精肝损伤模型组(P<0.05),且与正常对照差异不明显。 3.黑大豆种皮提取物低剂量组和高剂量组动物的肝脏脂肪病变程度,均明显低于酒精肝损伤模型组(P<0.05)。 结论: 黑大豆种皮提取物可以降低急性酒精性肝损伤动物的肝脏丙二醛(MDA)含量,防止肝组织还原型谷胱甘肽(GSH)的耗竭,改善急性酒精性肝损伤引起的肝脏病理组织学变化,,减轻肝脏脂肪变性的程度,有助于降低酒精性肝损伤。
[Abstract]:Black soybean not only has high nutritional value, but also contains many plant chemicals. Anthocyanin is the main bioactive substance of black soybean. Anthocyanins have many health effects such as anti-oxidation, anti-inflammation, improving vision, improving lipid metabolism, protecting cardiovascular system and so on. At present, there is no report on the effect of black soybean seed coat extract on alcohol-induced chemical liver injury. Objective: to determine the contents of reductive glutathione, malondialdehyde and triglyceride in the liver of mice with acute alcoholic liver injury. To investigate the effect of black soybean seed coat extract on alcoholic liver injury. Materials and methods: male Kunming mice were randomly divided into normal control group, alcohol model group, black soybean seed coat extract low dose group (30mg kg-1), black soybean seed coat extract high dose group (60mg kg-1). According to the Technical Specification for Health Food Inspection and Evaluation (2003 Edition), once a day, 31 days, 50% anhydrous ethanol solution was given once a day in addition to the normal control group, resulting in acute alcoholic liver injury. After fasting for 12 hours, the liver coefficient of each group was determined, and the content of GSH in the whole blood of MDA, liver, glutathione (GSH), MDA, liver, malondialdehyde (MDA) (MDA), brain tissue and liver triglyceride (TG),) were measured. The histopathological changes of liver were observed and hepatic fat lesion score was evaluated. Single factor ANOVA or rank sum test were performed with SPSS18.0 software. Results: 1. The content of GSH in liver of the high dose group of black soybean seed coat extract was significantly higher than that of the model group of alcoholic liver injury (P < 0. 05), and there was no significant difference between the high dose group and the normal control group (P < 0. 05). The content of GSH in liver tissue of the low dose group of black soybean peel extract was not significantly different from that of the model control group. 2. The content of MDA in liver tissue of low dose group and high dose group of black soybean peel extract was lower than that of alcohol induced liver injury group (P < 0. 05), and there was no significant difference compared with the normal control group (P < 0. 05). 3. The degree of hepatic fatty lesion in low dose and high dose groups of black soybean seed coat extract was significantly lower than that of alcohol induced liver injury model group (P < 0.05). Conclusion: black soybean seed coat extract can reduce malondialdehyde (MDA) content in liver and prevent the depletion of glutathione (GSH) in liver tissue. Improving the histopathological changes of liver caused by acute alcoholic liver injury and reducing the degree of hepatic steatosis are helpful to reduce alcoholic liver injury.
