性激素高效液相色谱检测新方法研究

【摘要】:性激素具有维持内分泌系统稳定的作用,监测其水平对哺乳动物疾病的预防、诊断、治疗具有极其重要的意义。目前,性激素在畜牧业生产中不规范使用,导致其在畜牧产品和环境水体中的残留。因此,建立准确、简便的性激素检测方法对保障人民生命健康和生态平衡尤为必要。本文研究了17β-雌二醇(BE2)、雌酮(E1)、己烯雌酚(DES)和黄体酮(PES)等性激素的高效液相色谱检测法,主要研究内容如下: 一、超声辅助表面活性剂增强乳化微萃取(UASEME)-高效液相色谱法检测性激素的研究 1.UASEME-HPLC检测环境水体中性激素的研究 以超声辅助表面活性剂增强乳化微萃取环境水中βE2、E1和DES,最佳萃取条件为:2.4x10-6mol/LTriton X-100作为分散剂和乳化剂,50μLCCl4为萃取剂,35kHz、25℃超声5min,4000rpm离心5min,富集倍数达85.29。10-1000ng/mL范围内,线性良好,检测限为0.10-0.20ng/mL,RSD为0.85-1.28%,回收率高于89.82%。此方法明显缩短了萃取时间,提高了富集倍数和萃取效率。 2. UASEME-HPLC检测血液、尿液和牛奶中性激素的研究 以超声辅助表面活性剂增强乳化微萃取生物和牛奶样品中βE2、E1、DES和PES,最佳萃取条件为:1mL乙腈和0.3g MgS04沉淀全血,0.2g Na2SO4沉淀尿液,0.1gNaAc、0.4g MgSO4口0.5mL乙腈重复两次沉淀牛奶。上清液调节pH至7.0,0.8×10-5mol/L Triton X-100为分散剂和乳化剂,80×L CCl4为萃取剂,35kHz、25℃下超声3min,加入0.3g NaCl,4000rpm离心5min,取有机相进样分析。5-1000ng/mL内,βE2、E1和DES线性良好,10-1000ng/mL范围内,PES的线性良好,检测限为0.05-0.50ng/mL,RSD为1.28-3.88%,回收率高于83.2%。此方法能准确、简便地分析血液、尿液和牛奶中的性激素,在临床和食品检测方面具有一定的推广性。 二、超声辅助浊点萃取(UA-CPE)—高效液相色谱法测定尿液中性激素的研究 以Tergitol TMN-6为萃取剂的浊点萃取尿液样品中βE2、E1和DES,最佳萃取条件为:0.5%(v/v)Tergitol TMN-6,35kHz、45℃超声45min,2.0%NaCl,4000rpm离心3min,富集倍数高于78.39。5-1000ng/mL范围内,线性良好,检测限为0.10-0.20ng/mL,回收率高于85.35%,重复检测三天的RSD为0.84-5.15%。此法有效地缩短了萃取时间,提高了萃取效率和稳定性。 三、盐析辅助液液萃取(SA-LLE)—高效液相色谱法检测牛奶中性激素的研究 以乙腈、NaAc和MgSO4盐析辅助液液萃取牛奶中BE2, El、DES和PES,最佳萃取条件为:15mL牛奶样品中,加入6mL酸化乙腈、g NaAc和2.4g MgSO4,10000rpm离心3min,取上清液重复离心一次后调节pH至5.0,加入1mL乙腈,4000rpm离心10min,取乙腈相进样分析。10-5000ng/mL范围内,BE2、E1和DES线性良好,20-8000ng/mL范围内,PES的线性良好,检测限为0.40-2.40ng/mL,RSD为0.3-4.4%,回收率高于95.10%,明显优于液液萃取和蛋白沉淀后再萃取。此法能有效地用于牛奶中性激素残留的检测。
