基于化学发光的卫生检测新方法研究

【摘要】:化学发光(Chemiluminescence, CL)检测法以其简单、成本低、分析速度快、灵敏度高等优点成为一种实用的微量及痕量分析方法。近年来,,发光试剂和体系不断得到改进与完善。随着新体系的开发以及与其它技术的联用,基于化学发光的流动注射技术和毛细管电泳的新方法以其快速、简便、灵敏等优点在卫生检验领域得到广泛应用。 本实验将化学发光检测技术与流动注射相结合,建立了测定注射液中盐酸罂粟碱的流动注射化学发光检测新方法;将鲁米诺-三价银(Ag(III))络合物化学发光新体系与毛细管电泳分离技术联用,建立了尿样中萘酚的分离及血清中精氨酸的分离新方法。 一、流动注射化学发光法测定盐酸罂粟碱注射液中的盐酸罂粟碱 目的:建立测定盐酸罂粟碱的流动注射化学发光新方法。 方法:碱性介质中,盐酸罂粟碱对鲁米诺-铁氰化钾化学发光体系有显著增强的作用,且在一定范围内,盐酸罂粟碱的浓度与化学发光强度成正比。基于此,本实验建立了流动注射-化学发光测定盐酸罂粟碱注射液中盐酸罂粟碱的方法,并研究了影响化学发光强度的因素。 结果:在最优条件下,测得最低检出限为1.70μg/mL,线性范围为5.00-200.00μg/mL (r=0.999),平均回收率为101.30%。 结论:该方法已用于盐酸罂粟碱注射液的测定,结果准确可靠。 二、毛细管电泳-化学发光法测定尿样中的1-萘酚和2-萘酚 目的:建立测定1-萘酚和2-萘酚的毛细管电泳化学发光新方法。 方法:研究发现,在碱性介质中,1-萘酚和2-萘酚对鲁米诺-Ag(III)络合物体系的化学发光有抑制作用。根据这一特点,建立了灵敏、快速的检测1-萘酚和2-萘酚的方法。通过对实验条件的优化,绘制标准曲线,将尿样处理后进行测定。 结果:在最优条件下,1-萘酚和2-萘酚的最低检出限分别为2.00×10~(-7)和6.00×10-6mol/L,相对标准偏差在2.03%-4.16%之间,平均加标回收率分别为101.50%和99.30%。 结论:该方法成功分离并测定实际尿样中的1-萘酚和2-萘酚,结果满意。 三、毛细管电泳-化学发光法测定血样中的精氨酸 目的:建立测定精氨酸的毛细管电泳化学发光新方法。 方法:在碱性介质中,精氨酸对鲁米诺-Ag(III)络合物体系的化学发光有增强作用。根据这一特点,建立了快速检测精氨酸的方法。通过对实验条件进行优化,绘制标准曲线,将血样处理后进行测定。 结果:在最优条件下,精氨酸的最低检出限为2.50×10~(-5)mol/L,相对标准偏差为1.75%,加标回收率为96.83%。 结论:该方法简单、快速,成功地用于血样中的精氨酸含量的检测。
[Abstract]:Chemiluminescent (Chemiluminescence, CL) detection has become a practical method for trace and trace analysis because of its advantages of simplicity, low cost, fast analysis and high sensitivity. In recent years, luminescent reagents and systems have been continuously improved and perfected. With the development of the new system and the combination with other technologies, the chemiluminescence-based flow injection technique and the new method of capillary electrophoresis have been widely used in the field of health examination for their advantages of fast, simple and sensitive. A new flow injection chemiluminescence detection method for the determination of papaverine hydrochloride in injection was established by combining chemiluminescence detection with flow injection. A new method for separation of naphthol from urine and separation of arginine from serum by using a new chemiluminescence system of luminol-trivalent silver (Ag (III) complex and capillary electrophoresis was established. 1. Flow injection chemiluminescence determination of papaverine hydrochloride in papaverine hydrochloride injection objective: to establish a new flow injection chemiluminescence method for the determination of papaverine hydrochloride. Methods: papaverine hydrochloride significantly enhanced the chemiluminescence system of luminol-potassium ferricyanide in alkaline medium, and the concentration of papaverine hydrochloride was proportional to the intensity of chemiluminescence in a certain range. Based on this, a flow injection-chemiluminescence method for the determination of papaverine hydrochloride in papaverine hydrochloride injection was established, and the factors affecting the intensity of chemiluminescence were studied. Results: under the optimum conditions, the detection limit was 1.70 渭 g / mL, the linear range was 5.00-2000.00 渭 g / mL (r = 0.999), and the average recovery was 101.30%. Conclusion: the method has been applied to the determination of papaverine hydrochloride injection with accurate and reliable results. Determination of 1-naphthol and 2-naphthol in urine by capillary electrophoresis-chemiluminescence method objective: to establish a new capillary electrophoresis chemiluminescence method for the determination of 1-naphthol and 2-naphthol. Methods: it was found that in alkaline medium, 1-naphthol and 2-naphthol inhibited chemiluminescence of Lumino-Ag (III) complex system. A sensitive and rapid method for the determination of 1-naphthol and 2-naphthol was established. By optimizing the experimental conditions, the standard curve was drawn, and the urine samples were measured after treatment. Results: under the optimum conditions, the detection limits of 1-naphthol and 2-naphthol were 2.00 脳 10 ~ (- 7) and 6.00 脳 10 ~ (- 6) mol / L, respectively. The relative standard deviation (RSD) was 2.03% ~ 4.16%. The average recoveries were 101.50% and 99.30%, respectively. Conclusion: the method is satisfactory for the separation and determination of 1-naphthol and 2-naphthol in actual urine samples. 3. Determination of Arginine in Blood samples by Capillary Electrophoresis-Chemiluminescence method objective: to establish a new chemiluminescence method for determination of arginine by capillary electrophoresis. Methods: arginine enhanced chemiluminescence of Lumino-Ag (III) complex system in alkaline medium. According to this characteristic, a rapid method for the determination of arginine was established. The experimental conditions were optimized, the standard curve was drawn, and the blood samples were measured. Results: under the optimum conditions, the detection limit of arginine was 2.50 脳 10 ~ (- 5) mol/L, relative standard deviation was 1.75%, and the recovery rate was 96.83%. Conclusion: the method is simple, rapid and successful for the determination of arginine in blood samples.
