RANKL-三氯乙烯致大鼠毒性损伤的关键因子

【摘要】:三氯乙烯(Trichloroethylene,TCE),是一种氯代溶剂,由于其具有良好的亲脂性和低熔点的物理特性,在工业生产中是一种理想的去油剂和干燥剂,在家庭生活中,广泛应用于粘合剂,润滑剂和改正液等产品中。但由于在生产生活中使用TCE后未妥善处理,使得越来越多的TCE进入环境当中,成为一种环境污染物,危害人类的健康。TCE具有多种作用,包括神经毒性,肾损伤,肝损伤,肺损伤,心率变化,免疫器官病理变化等,然而,迄今为止,TCE对机体的毒性作用机制还不清楚。因此,TCE已被列入美国国家环保局在1976年公布的129种优先污染物和我国环境特征污染物的“黑名单”。 核因子-κB受体活化因子配体(Receptor Activator for Nuclear Factor-κBLigand,RANKL)属于肿瘤坏死因子家族的成员,其受体是核因子κB受体活化因子(receptor activator of NF-κB,,RANK)。RANKL具有多种作用,包括参与骨重建,淋巴结的形成,胸腺微环境的建立,怀孕过程中的乳腺发育,激素引起的乳腺癌,和癌症过程中的骨转移等过程。 本文通过灌胃给予TCE致大鼠心,肝,肺,肾,脾和胸腺的损伤,采用苏木精-伊红染色、免疫组织化学和Western-Blot方法检测了TCE对大鼠毒性损害及RANKL的表达情况,系统性的研究了TCE引起的毒性损伤及RANKL之间的相关性。结果发现TCE能明显的引起大鼠体重的下降,加剧各器官的组织病理学形态损伤,且能提高不同组织中RANKL的表达,表明RANKL参与了的TCE对机体损伤。此研究为环境毒素TCE的毒性及其作用机制提供理论基础。
[Abstract]:Trichloroethylene (Trichloroethylene,TCE) is a kind of chlorine solvent. It is an ideal deoiling agent and desiccant in industrial production because of its good lipophilicity and low melting point physical properties. It is widely used in family life. Lubricant and correction liquid and other products. However, due to the lack of proper treatment after the use of TCE in production and life, more and more TCE enter the environment and become an environmental pollutant, which endangers human health. TCE has many functions, including neurotoxicity, kidney injury and liver injury. Lung injury, heart rate changes, pathological changes of immune organs, etc. However, so far, the toxic mechanism of TCE on the body is not clear. Therefore, TCE has been listed in the blacklist of 129 priority pollutants and environmental characteristic pollutants published by the National Environmental Protection Agency in 1976. Nuclear factor-kappa B receptor activating factor ligand (Receptor Activator for Nuclear Factor- 魏 BLigand, RANKL) belongs to tumor necrosis factor family. Its receptor is nuclear factor kappa B receptor activating factor (receptor activator of NF- kappa B, RANK). Rankl has many effects. It includes bone reconstruction, lymph node formation, thymic microenvironment establishment, breast development during pregnancy, hormone-induced breast cancer, and bone metastasis during cancer. The injury of heart, liver, lung, kidney, spleen and thymic gland induced by TCE was detected by hematoxylin-eosin staining, immunohistochemistry and Western-Blot. The toxic damage and the expression of RANKL in rats were detected by TCE. The correlation between toxic damage induced by TCE and RANKL was studied systematically. The results showed that TCE could significantly decrease the body weight, aggravate the pathological morphological damage of various organs, and increase the expression of RANKL in different tissues, indicating that RANKL was involved in the injury of TCE to the body. This study provides a theoretical basis for the toxicity and mechanism of environmental toxin TCE.
