壬基酚对雄性小鼠生精能力及睾酮的影响

【摘要】:[目的]研究壬基酚对雄性小鼠生精能力及血清性激素睾酮的影响。[方法]雄性昆明种小鼠40只,随机分为4组,每组10只,分别为壬基酚25、50、100 mg/kg剂量染毒组和溶剂对照组(玉米油)。各组隔日灌胃给予相应剂量的壬基酚溶液,连续35 d后处死小鼠取血,用酶联免疫法测定小鼠血清睾酮的水平。取左侧睾丸做组织病理切片,取双侧附睾制备精子悬液,并在光学显微镜下进行精子计数,计算活精子率和精子畸形率。[结果]35 d后,与对照组相比,100 mg/kg剂量染毒组小鼠睾丸组织病理学切片显示曲细精管萎缩,体重减轻幅度升高,睾丸重量下降,精子数量、精子活动度均降低,精子畸形率增高(均P0.05)。染毒组小鼠血清睾酮含量、精子数量与壬基酚浓度存在明显剂量-反应关系(r=-0.98,r=-0.96;P0.05)。[结论]壬基酚对雄性小鼠生殖系统有一定损害作用,高剂量的壬基酚能损伤小鼠的生精功能。
[Abstract]:Objective to study the effect of nonylphenol on spermatogenesis and serum sex hormone testosterone in male mice. [methods] 40 male Kunming mice were randomly divided into 4 groups with 10 mice in each group: nonylphenol 25, 50100 mg/kg dose group and solvent control group (corn oil). The mice in each group were given the corresponding dose of nonylphenol solution intragastrically every other day. After 35 days, the mice were killed and the serum testosterone level was measured by enzyme-linked immunosorbent assay (Elisa). The left testes were taken as pathological sections, the bilateral epididymis was taken to prepare sperm suspension, and the sperm count was carried out under optical microscope to calculate the live sperm rate and sperm deformity rate. [results] after 35 days, compared with the control group, the pathological sections of testicular histology in the 100 mg/kg dose group showed that the seminal tubules were atrophied, the weight loss was increased, the testicular weight was decreased, the sperm count and sperm motility were decreased, and the sperm deformity rate was increased (P 0.05).
