模拟地震应激对大鼠心理行为与神经内分泌影响的研究

【摘要】:背景 地震被称为群灾之首,其发生的频率高、破坏性极强,经历地震的人们身心健康可能受到重创。目前强震巨灾后的心理问题日益引起社会的广泛重视,针对不同的心理问题目前存在不同的干预手段,音乐疗法作为一种新兴的治疗方式目前在临床不同科室中得到应用,并且已取得了一定的成效。然而音乐疗法在强震巨灾后的心理问题治疗方面鲜有报道。目前科研工作者对音乐治疗的研究主要集中于临床观察研究,还缺少详实的动物实验和更为先进的指标。通过动物实验来寻找音乐治疗对疾病病理生理的影响机制,从而为音乐治疗提供一定的实验室理论依据。 目的 研究模拟地震应激对大鼠心理行为、血清CORT及脑组织中GR、MR的影响作用,同时探讨早期音乐干预能否有效改善模拟地震应激对大鼠影响,有望为未来灾害现场的早期干预提供理论依据。 方法 1.应用已经与北京维尔森科技发展有限公司成功研制的模拟地震震动平台,可以模拟地震发生的环境。10周龄雄性Wistar大鼠30只,体重200g-230g,30只按体重随机分为如下三组,C组(control)10只未经历模拟地震刺激的实验室内正常的大鼠;S组(stress):10只大鼠,给予慢性模拟地震刺激30min/d,持续3w;M组(stress+music):10只大鼠,给予慢性模拟地震刺激30min/d,同时予音乐治疗3h/d,持续3w。 2.模拟地震刺激结束后分别于第一天、第二天行旷场行为学检测、悬尾实验检测,于第三天活杀各组大鼠,检测血清CORT,取脑组织,对海马、额叶皮层部位进行HE常规病理切片,并行免疫组织化学法观察MR、GR的表达。 结果 经历三周模拟地震应激后,大鼠血清CORT较正常对照组明显升高,差异具有统计学意义(P0.05),存在明确的情绪行为改变,主要表现为情绪的焦虑和抑郁(P0.05),海马、额叶皮层中MR、GR表达显著下调(P0.05)。在给予模拟地震应激的同时进行早期音乐干预,大鼠血清CORT较正常对照组显著下降(P0.05),MR、GR的表达明显上调(P0.05),而大鼠行为学的改变未达到显著性差异(P0.05)。 结论 1.慢性模拟地震应激可以导致大鼠出现焦虑、抑郁情绪,血清CORT水平升高,海马、额叶皮层中皮质酮受体的表达下调。 2.应激引起的CORT升高对海马、额叶皮层神经元具有一定的细胞毒性作用。 3.早期合理音乐干预治疗可拮抗模拟地震应激对大鼠血清CORT的升高作用,对大鼠行为的改变有部分改善作用。
[Abstract]:Background earthquakes are called the first mass disasters, their frequency is high and destructive, and the physical and mental health of people who have experienced earthquakes may be severely damaged. At present, the psychological problems after strong earthquake catastrophe have attracted more and more attention of the society. In view of different psychological problems, there are different intervention methods at present. Music therapy, as a new treatment method, has been applied in different clinical departments, and has achieved some results. However, music therapy is rarely reported in the treatment of psychological problems after strong earthquake catastrophe. At present, the research of music therapy is mainly focused on clinical observation and research, and there is still a lack of detailed animal experiments and more advanced indicators. Through animal experiments, we can find out the mechanism of the influence of music therapy on the pathophysiology of diseases, so as to provide a certain laboratory theoretical basis for music therapy. Objective to study the effects of simulated earthquake stress on psychological behavior, serum CORT and GR,MR in rats, and to explore whether early music intervention can effectively improve the effect of simulated earthquake stress on rats, which is expected to provide theoretical basis for early intervention in disaster sites in the future.
