有氧运动联合“消脂散”对高脂膳食大鼠胰岛素抵抗的干预作用及其机制的研究

【摘要】:实验目的: 探讨有氧运动联合“消脂散”对高脂膳食大鼠胰岛素抵抗的干预作用及作用机制 试验方法: 实验大鼠分为5组:普通饲料对照组(C)、高脂膳食组(HF)、高脂膳食+有氧运动组(HE)、高脂膳食+消脂散组(HZ)、高脂膳食+有氧运动+消脂散组(HZE)。运动方案为10周60min无负重游泳运动,每周6天。消脂散灌胃为lml/l100gBW剂量。试验结束后用相关试剂盒测定空腹血糖(FPG)、空腹胰岛素(FINS)、甘油三脂(TG)、总胆固醇(TC)、游离脂肪酸(FFA)、血清肿瘤坏死因子-α(TNF-α)、瘦素(leptin)等指标,用实时荧光定量检测大鼠肝脏中固醇调节原件结合蛋白-1c (SREBP-1c) mRNA和细胞因子信号传导抑制因子-3(SOCS-3) mRNA的表达量,并计算胰岛素敏感性指数(ISI)、胰岛素抵抗指数(HOMA-IR)来帮助判断IR的形成。 结果: 1与C组相比,HF组大鼠FPG、INS、HOMA-IR、TG、TC、FFA、leptin和TNF-α均显著提高(P0.01,P0.05),ISI显著降低(P0.05)。通过相关性分析,TG、 TC、FFA、leptin和TNF-α与HOMA-IR和ISI具有显著的相关性(P0.05),提示高脂膳食可以通过不同途径导致IR的形成; 2通过双因素方差分析,有氧运动或者消脂散均能降低FPG、FINS、HOMA-IR、TG、 leptin和TNF-α的含量(P0.01,P0.05),显著提高ISI(P0.05);另外,有氧运动具有降低TC和FFA的趋势(P0.05),而消脂散则可以显著降低其含量(P0.05),提示有氧运动或者消脂散均能通过不同途径改善高脂引起的IR; 3通过双因素方差分析,有氧运动和消脂散对FPG、FINS、HOMA-IR、ISI、TG、 TC、FFA、leptin和TNF-α均具有显著的交互作用(P0.05),提示有氧运动联合消脂散比单独有氧运动干预或消脂散干预具有更好的改善IR的作用; 4通过实时荧光定量PCR和双因素方差分析,与C组相比,高脂膳食会上调肝脏组织SOCS-3和SREBP-1c的表达(P0.01),而有氧运动或消脂散则能抑制其表达(P0.01,P0.05),并且二者具有很好的交互作用(P0.05)。通过相关性分析,FPG、 FINS、HOMA-IR、ISI、TG、TC、FFA、leptin和TNF-α均与SOCS-3和SREBP-1c且有显著的相关性(P0.05),提示有氧运动联合消脂散可能是通过下调肝脏SOCS-3和SREBP-1c的表达而调节FPG、FINS、TG、TC、FFA、leptin和TNF-α水平而起到改善IR的作用。 结论: 1高脂膳食会引起大鼠脂质代谢紊乱和leptin、TNF-α水平升高,引起IR; 2有氧运动和消脂散可以改善大鼠脂质代谢紊乱和降低leptin、TNF-α水平,起到预防IR的作用,并且有氧运动和消脂散具有显著的交互作用; 3高脂膳食会引起大鼠肝脏SOCS-3和SREBP-1c基因表达的增加,而有氧运动和消脂散则可以抑制其表达,并且二者具有显著的交互作用,这也可能是有氧运动和消脂散预防IR的分子机制。
[Abstract]:Objective: Effect of aerobic exercise combined with Xiaozhi Powder on Insulin Resistance in Rats with High Fat Diet and its Mechanism Test methods: Rats were divided into 5 groups: normal diet control group, high fat diet aerobic exercise group, high fat diet Xiaozhi powder group, high fat diet aerobic exercise dissipating powder group. The exercise program was 10 weeks 60min swimming without load, 6 days a week. The dosage of Xiaozhi San was lml/l100gBW. At the end of the experiment, fasting blood glucose (FPGN), fasting insulin (FINSN), triglyceride (TGG), total cholesterol (TC), free fatty acid (FFAA), serum tumor necrosis factor- 伪 (TNF- 伪), leptin (leptinin) were measured with relevant kit. The expression of steroid regulatory original binding protein (SREBP-1c) mRNA and cytokine signal transduction suppressor factor (-3) SOCS-3 (mRNA) in rat liver was detected by real-time fluorescence quantitative analysis. The insulin sensitivity index (ISI) and insulin resistance index (HOMA-IRI) were calculated to help judge the formation of IR. Results: 1 compared with group C, both FFAleptin and TNF- 伪 of FFAleptin and TNF- 伪 were significantly increased in FFAleptin and TNF- 伪. There was a significant correlation between TG, TCG-FFA-leptin and TNF- 伪 and HOMA-IR and ISI, suggesting that high fat diet could lead to the formation of IR in different ways. 2By the analysis of variance of two factors, aerobic exercise or Xiaozhi San could decrease the contents of TG, leptin and TNF- 伪 in FINSHOMA-IRN, and increase the content of Igna, P0.05, P0.05, in addition, the contents of FINSHOMA-IRG, TNF- 伪 and TNF- 伪 were significantly increased. Aerobic exercise has the tendency of decreasing TC and FFA, while Xiaozhi Powder can significantly reduce the content of P0.05N, suggesting that aerobic exercise or Xiaozhi Powder can improve IRI caused by high fat through different ways. (3) based on the analysis of variance of two factors, aerobic exercise and Xiaozhi Powder had significant interaction on FINSHOMA-IRISII TG, TCU FFAFAleptin and TNF- 伪, suggesting that aerobic exercise combined with Xiaozhi Powder had a better effect on improving IR than aerobic exercise alone or Xiaozhisan intervention. 4 by real-time fluorescence quantitative PCR and double factor analysis of variance, compared with group C, high-fat diet could up-regulate the expression of SOCS-3 and SREBP-1c in liver tissue, while aerobic exercise or Xiaozhi San could inhibit the expression of P0.01and P0.05, and had a good interaction between the two groups. There was a significant correlation between SOCS-3 and TNF- 伪 and SOCS-3 and TNF- 伪, suggesting that aerobic exercise combined with Xiaozhi Powder could improve IR by down-regulating the expression of SOCS-3 and SREBP-1c in the liver and regulating the levels of FFAleptin and TNF- 伪 in the liver. Conclusion: 1High fat diet could induce lipid metabolism disorder and increase the level of leptinine TNF- 伪 in rats. 2aerobic exercise and Xiaozhi Powder can improve the disorder of lipid metabolism and decrease the level of leptinine TNF- 伪, and play a role in preventing IR, and aerobic exercise and Xiaozhi Powder have significant interaction; 3High fat diet could increase the expression of SOCS-3 and SREBP-1c genes in the liver of rats, but aerobic exercise and Xiaozhi Powder could inhibit their expression, and they had significant interaction, which may also be the molecular mechanism of aerobic exercise and Xiaozhi Powder to prevent IR.
