双白术内酯对Aβ 25-35 损伤神经细胞的保护及其与mPTP开放的研究

目的研究白术中双白术内酯对Aβ淀粉样蛋白(Aβ25-35)神经损伤的大鼠肾上腺嗜铬细胞瘤细胞(PC12)及人骨髓神经母细胞瘤细胞系(SH-SY5Y)诱导其线粒体膜通透性转换孔(mPTP)的开放和细胞凋亡的的保护作用及其机制研究,并进一步提供基于药代动力学和药理学数据的系统网络药理学模型,以确定中药白术的有效化合物和潜在靶点,为白术对阿尔茨海默病具有神经保护作用提供理论基础。方法体外培养PC12和SH-SY5Y细胞,用20μMAβ25-35处理细胞构建细胞损伤模型,对照组用不同浓度双白术内酯预处理2小时。实验组如下:(1)空白对照组,(2)Aβ25-35损伤组,(3)药物对照组(低,中,高)。通过MTT法检测细胞增殖;用罗丹明123染色检测线粒体膜电位;ROS试剂盒检测细胞内ROS;蛋白质印迹分析检测线粒体相关蛋白CypD和CytC的变化。网络药理学能构建生物网络并分析网络中成分、靶点和疾病之间的联系。我们通过使用Cytoscape 3.2.1软件构建了以下网络图:(1)C-T(成分-靶点图),在TCMSP和SWISS数据...
