黄芩苷纳米脂质体抑制TLR4信号通路减轻小鼠非酒精性脂肪肝病

  背景:非酒精性脂肪肝病(non-alcoholic fatty liver disease,NAFLD)是以肝实质细胞脂肪变性、脂肪贮积和炎症反应为主要特征,并且无过量饮酒史的慢性肝脏代谢性疾病。NAFLD从单纯性脂肪肝、可发展成为非酒精性脂肪性肝炎(non-alcoholic steatohepatitis,NASH)、肝纤维化、肝硬化、甚至肝癌。NAFLD的发病率、死亡率逐年升高,并呈低龄化发病趋势。尽管目前对NAFLD发病机制的研究取得了一定的进展,但仍然缺乏有效的治疗药物。因此,寻找对NAFLD有效的治疗药物就显得尤其关键。黄芩苷(Baicalin,BA)是一种具有抗炎、抗氧化、调血脂、免疫调节等多种药理作用的黄酮类化合物。研究发现黄芩苷对多种原因引起的肝脏损伤有一定的保护作用。目的:探讨黄芩苷及其纳米脂质体对小鼠非酒精性脂肪肝病的保护作用及可能的作用机制。方法:1.黄芩苷纳米脂质体的制备及表征鉴定以卵磷脂、胆固醇为膜材料,采用薄膜分散法制备黄芩苷纳米脂质体,并考察其微观形态、粒径分布、多分散系数、Zeta电位值、包封率和载药量。2.黄芩苷纳米脂质体对NAFLD小鼠的保护作用将...
