蜈蚣提取物对人肝癌细胞HepG2及肝癌原位移植瘤STAT3信号通路的影响(英文)

  目的通过观察蜈蚣提取物(CSE)对人肝癌细胞Hep G2及裸鼠肝癌原位移植瘤的影响,探讨其抗肝癌的机制。方法 Hep G2细胞分为CSE250(250μg/m L)处理组、CSE500(500μg/m L)处理组、5-FU(5-氟尿嘧啶)处理组和对照组,采用酶解丙酮沉淀法分离纯化CSE并作用于人肝癌细胞HepG2,CCK8法检测细胞增殖抑制,流式细胞术检测细胞周期变化。Western blot检测CSE处理后HepG2细胞信号传导与转录激活因子3(STAT3)信号通路相关蛋白表达变化。制备人肝癌细胞裸鼠原位移植瘤模型,随机分为模型组、5-FU组、CSE10组[10mg/(kg·d)]和CSE50组[50mg/(kg·d)],每组12只,观察裸鼠原位移植瘤瘤体体积及质量变化,并检测瘤体p-STAT3、p38丝裂原激活的蛋白激酶(p38MAPK)蛋白表达水平。取模型组及CSE50组裸鼠肝原位移植瘤组织,采用蛋白酪氨酸激酶(PTKs)芯片技术检测并对差异蛋白进行GO及KEGG分析。结...
