【摘要】:目的观察电针及侧脑室微量注射腺苷A1受体激动剂(CCPA)和拮抗剂(DPCPX)对弗氏完全佐剂诱发的炎症痛模型(Adjuvant Arthritis,简称AA)大鼠的镇痛和免疫调节作用,以探讨腺苷A1受体参与炎症痛及电针的镇痛抗炎作用机制。观察腺苷A1受体、5-HT1A受体、鸟苷酸结合蛋白、丝裂原活化蛋白激酶在AA大鼠下丘脑和脊髓的表达,初步探讨相关受体和信号转导通路参与电针镇痛抗炎的中枢机制。 方法:以弗氏完全佐剂诱发的炎症痛模型(AA)大鼠为研究对象,采用辐射热行为学测痛法、排水容积法、酶联免疫吸附法(ELISA),观察电针腰夹脊穴和侧脑室微量注射CCPA、DPCPX对AA大鼠痛阈、足跖容积和血清IL-1β、IL-12的影响;应用免疫组织化学技术法、实时荧光定量PCR等方法,观察下丘脑和脊髓腺苷A1受体、5-HT1A受体、G蛋白、MAPK的表达及电针的影响。 结果: 1、电针夹脊穴可提高AA大鼠痛阈,侧脑室微量注射CCPA可提高AA大鼠痛阈并加强电针的镇痛作用,DPCPX则降低AA大鼠痛阈同时逆转电针的镇痛作用; 2、电针夹脊穴和侧脑室注射CCPA可降低AA大鼠血清IL-1β、IL-12水平,DPCPX则升高IL-1β、IL-12水平并逆转电针的抗炎作用; 3、AA大鼠下丘脑和脊髓ADORA1表达下调,电针可上调该受体表达,CCPA上调其表达且与电针具有一定的协同作用,DPCPX则抑制ADORA1表达,并抑制电针的作用; 4、AA大鼠下丘脑和脊髓5-HT1AR表达上调,电针可下调5-HT1AR表达; 5、AA大鼠下丘脑和脊髓GimRNA表达上调GsmRNA表达下调,电针可下调Gi mRNA表达并上调GsmRNA表达;AA大鼠下丘脑和脊髓P38和JNKmRNA表达上调,电针则下调二者基因表达。 结论:ADORA1参与炎症痛和电针镇痛,是电针镇痛抗炎作用的重要靶点之一;电针镇痛的中枢作用机制与ADORA1、5-HT1AR及Gi/Gs蛋白、P38, JNK介导的信号转导通路有关。
[Abstract]:Objective To observe the effect of electroacupuncture and intracerebroventricular injection of adenosine A1 receptor agonist (CCPA) and antagonist (DPCPX) on the model of inflammatory pain induced by Freund's complete adjuvant (Adjuvant Arthritis, referred to as AA) Regulation of analgesia and immunity in rats, to explore the mechanism of analgesic and anti-inflammatory effects of adenosine A1 receptors are involved in inflammatory pain and EA. Adenosine A1 receptor, 5-HT1A receptor, GTP binding protein, expression of mitogen activated protein kinase in the hypothalamus and spinal cord of AA rats, to investigate the central mechanism of related receptors and signal transduction pathways involved in the EA analgesic and anti-inflammatory.
Methods: using Freund's complete adjuvant induced inflammatory pain model (AA) rats were studied by thermal radiation behavior pain measuring method, displacement method, enzyme-linked immunosorbent assay (ELISA), to observe the effects of microinjection of CCPA into the waist clip ridge cave and the lateral ventricle, DPCPX on pain threshold in AA rats, foot paw volume and serum IL-1 beta, IL-12; immunohistochemistry method, real-time fluorescence quantitative PCR, receptor, hypothalamus and spinal cord adenosine A1 on 5-HT1A receptor, G protein, expression and effect of Electroacupuncture of MAPK.
