乳腺癌差异基因筛选及PAIP1对其生物学行为的影响

研究背景:乳腺癌是源于乳腺导管上皮的恶性肿瘤,近年来其发病率呈上升的趋势,已成为发病率最高的妇科肿瘤。现已公认,转移和复发是导致乳腺癌患者死亡的主要原因,因此,明确乳腺癌的转移机制、发现有效的治疗靶点是治疗乳腺癌的关键。研究目的:应用基因芯片技术和高内涵筛选(High Contents Screening, HCS)等较为先进的分子生物学技术明确乳腺癌的差异表达基因,继而通过体外功能实验初步探讨这些差异基因在乳腺癌演进过程中的作用及可能的分子机制,通过体内实验验证差异基因蛋白表达,检测在乳腺癌预后评估及治疗模式风险预测中的临床应用价值,为乳腺癌提供有效的预后评估新分子标志物及治疗靶点。材料与方法:1)应用基因芯片技术检测20对乳腺癌与正常乳腺组织中的基因表达谱,初步筛选差异表达基因,继而应用实时荧光定量PCR (Quantitative Real-time PCR, qRT-PCR)和HCS等生物学技术明确乳腺癌的差异表达基因;2)制备慢病毒PAIP1 shRNA质粒,沉默MDA-MB-231乳腺癌细胞中的目的基因,继而应用荧光标记显微镜观察计数以及流式细胞技术等检测细胞增殖、凋亡以及细...
