丰富环境和游泳运动对自闭症大鼠CX43、GSK3β、Bcl-2蛋白表达的影响

【摘要】:目的:观察丰富环境和游泳干预对自闭症模型大鼠脑皮CX43、GSK3β、Bcl-2蛋白表达的调控。实验方法:将雌、雄Wistar大鼠各1只置于同一笼内并过夜,当检测到雌鼠排出在便盘或者一直留在阴道内的淡黄色或白色的半透明的阴栓记胚胎为第0.5天。将受孕成功的母鼠挑出单独喂养,并随机将受孕雌鼠分成2组,一组使用0.85%的生理盐水配VPA粉沫并制作成250mg/ml溶液,在胚胎12.5天的时候将该溶液注入受孕母鼠体内(600mg/kg),该组受孕母鼠生产下的仔鼠记录为模型组,对模型组的仔鼠进行筛选,得到自闭症模型大鼠;对照组是未进行任何干预的正常母鼠产下的正常仔鼠。本论文实验分为五组:游泳+丰富环境组(SE+EE)、丰富环境组(EE)、自闭症组(ASD)、游泳运动组(SE)、空白对照组(C)。游泳运动组于每天下午3:00——4:30在30±1℃的水中进行持续90分钟的中等负荷运动训练,并持续训练8周。丰富环境组生活在长、宽、高为1m×0.6m×0.4m的透明饲养笼中,笼内有多种玩具,让自闭症大鼠在其中训练。干预结束后,断头取脑,用免疫组化方法检测大鼠脑皮层CX43、GSK3β、Bcl-2蛋白的表达程度,应用Image-Pro Plus多功能真彩色细胞图像分析管理系统进行图像采集分析,同时计算出CX43、GSK3β、Bcl-2蛋白的平均荧光强度(荧光强度/总面积),并做出对比分析。实验所得数据用SPSS13.0统计软件进行分析。实验结果:1.体重对比:与正常组大鼠体重相比,VPA自闭症模型组仔鼠在出生后的第7天、第14天、第21天,PN70.05,有明显的不同;PN14和PN21均0.05,没有明显的不同。2.睁眼时间:与正常组仔鼠相比,VPA模型组仔鼠的睁眼时间要稍微晚一些,PN13、PN14和PN15天时,在统计学上都具有差异性(P0.05)。3.方向趋向性:自闭症幼鼠较正常组幼鼠相比,其转身180o所用的时间要长一些,在PN7、PN8和PN9天,P0.05,在统计学上都有差异性。4.强迫游泳:与正常大鼠相比较,在PN8、PN10天,自闭症模型组大鼠的游泳能力明显要弱,P≤0.05,具有统计学差异。5.CX43蛋白表达:与模型组比较,游泳+丰富环境组脑皮质CX43蛋白表达较多,P0.05,具有显著差异;与游泳运动组比较,丰富环境组CX43蛋白表达多于游泳组,P0.05,具有显著差异。6.GSK3β蛋白表达:与模型组比较,游泳+丰富环境组GSK3β蛋白表达多于模型组,P0.05,具有显著差异;与丰富环境组比较,游泳组脑皮质GSK3β蛋白表达较多,P0.05,有统计学差异。7.Bcl-2蛋白表达:与模型组比较,游泳+丰富环境组脑皮质Bcl-2蛋白表达较少,P0.05,具有显著差异;与游泳运动组比较,丰富环境组Bcl-2蛋白表达P0.05,没有统计学差异。结论:丰富环境和游泳运动可以增加CX43、GSK3β蛋白在自闭症模型大鼠脑皮层的表达,降低Bcl-2蛋白的表达。
[Abstract]:Objective: to observe the regulation of rich environment and swimming intervention on the expression of Bcl 2 protein of CX43 (GSK3 尾) Bcl 2 in rat cortex. Methods: one male and one male Wistar rats were placed in the same cage and overnight. When a light yellow or white translucent vaginal suppository was detected in the feces or in the vagina, the embryo was marked at day 0.5. The successful pregnant rats were singled out for feeding alone and were randomly divided into two groups. One group used 0.85% normal saline and VPA powder foam to make 250mg / ml solution, and injected the solution into the pregnant mice at 12.5 days after embryo. The offspring produced by the group were recorded as model group, and the offspring of the model group were screened. The rat model of autism was obtained. The control group was a normal newborn rat without any intervention.
