肥胖SD大鼠运动性减肥与白色脂肪细胞棕色化的研究

【摘要】:观察有氧运动对肥胖SD大鼠白色脂肪棕色化相关细胞因子差异表达的影响,并探讨白色脂肪棕色化的分子机制。方法:将建模成功的肥胖SD大鼠分为运动组和肥胖组。运动组大鼠进行8周的跑台运动,肥胖组和对照组不给予运动。取各组大鼠皮下白色脂肪和肩胛下棕色脂肪,采用肉眼观察和镜下观察;用Real time-PCR检测各组脂肪组织中UCP-1、PGC-1α、Irisin和PRDM16 mRNA表达水平;用Western blot检测各组脂肪组织中UCP-1蛋白表达。结果:1)通过肉眼观察到运动组SD大鼠腹股沟皮下白色脂肪组织中夹杂着数块黄豆大浅棕色的脂肪组织,光镜下类似棕色脂肪细胞;Real time-PCR检测显示UCP-1、PGC-1α和irisin mRNA的表达水平明显高于肥胖组和对照组皮下白色脂肪细胞,Western blot显示UCP-1蛋白表达水平明显高于对照组和肥胖组皮下白色脂肪细胞。2)运动组SD大鼠肩胛间的棕色脂肪组织肉眼观察平均体积变大,弹性好,镜下血管丰富,检测UCP-1、PGC-1α、irisin mRNA表达量明显高于肥胖组和对照组的棕色脂肪组织。3)运动组SD大鼠的腹腔及肾周的白色脂肪经仔细检查和取材均未发现浅棕色改变。结论:1)有氧运动可促进肥胖SD大鼠白色脂肪细胞棕色化;2)有氧运动提高了SD大鼠肩胛间棕色脂肪的功能,促进了细胞分化;3)有氧运动对SD大鼠的腹腔及腹膜后的白色脂肪棕色化的作用微弱,不同部位的白色脂肪可能来源不同,导致细胞构成和细胞功能也不同。
[Abstract]:To observe the effect of aerobic exercise on the differential expression of cytokines associated with white fat browning in obese SD rats and to explore the molecular mechanism of white fat browning.Methods: obese SD rats were divided into exercise group and obese group.Rats in exercise group were given treadmill exercise for 8 weeks, while obese group and control group were not given exercise.The subcutaneous white fat and subscapular brown fat were taken from the rats in each group, the expression of UCP-1PGC-1 伪 -Irisin and PRDM16 mRNA in adipose tissue was detected by Real time-PCR, and the expression of UCP-1 protein in adipose tissue was detected by Western blot.Results the white fat tissue of groin subcutaneous of SD rats in exercise group was observed by naked eye.The expression levels of UCP-1PGC-1 伪 and irisin mRNA in brown adipocytes were significantly higher than those in obese group and control group. Western blot showed that UCP-1 protein expression level was significantly higher than that in control group and obese group.The average volume of brown adipose tissue between scapula of SD rats was larger with naked eyes.The expression of UCP-1PGC-1 伪 nirisin mRNA was significantly higher than that in obese group and control group. 3) the white fat in abdominal cavity and perirenal of SD rats in exercise group showed no light brown color changes after careful examination and sampling.Conclusion aerobic exercise can promote the browning of white adipocytes in obese SD rats.) aerobic exercise can improve the function of interscapular brown fat in SD rats.The effect of aerobic exercise on white fat browning in abdominal cavity and retroperitoneum of SD rats was weak, and the white fat in different parts might come from different sources, resulting in different cell composition and cell function.
