两种肌肉工作形式对大鼠脂代谢调节肌肉因子Irisin、CTRP15的影响

【摘要】:研究目的:运动诱导骨骼肌分泌脂代谢调节肌肉因子,进而促进脂肪氧化燃烧是近年来运动减肥机制的研究热点,本实验探讨相同摄氧量下,两种肌肉工作形式(向心运动和离心运动)运动后对血清Irisin、CTRP15的不同影响,以揭示相同能量代谢水平下,不同运动形式对脂肪氧化调控的区别。研究方法:研究选用纯系雄性Sprague-Dawley(SD)大鼠90只,8周龄,体重200±20g。大鼠随机分为对照组(n=6)、平坡跑运动组(n=30)和下坡跑运动组(n=30)。运动组的运动时间都为60分钟,下坡跑运动组的坡度为-5°。在运动结束后的0h、6h、12h、24h和48h每组取6只大鼠分别进行心脏取血、腓肠肌和比目鱼肌取材,检测各时间点FNDC5水平。采用ELISA方法检测大鼠血清Irisin水平和CTRP15的水平。研究结果:(1)运动时间相同,跑速有别情况下,大鼠平坡跑和下坡跑的摄氧量水平可以无统计学差异(P0.05)。(2)向心运动组和离心运动组大鼠运动后6h,腓肠肌FNDC5的表达水平显著增加(P0.05);向心运动组运动后0h、12h、24h、48h与对照组相比,没有显著性差异;离心运动组运动后0h、12h、24h、48h与对照组相比,显著提高。(3)大鼠运动后血清Irisin水平的比较:向心运动组与对照组相比,运动后0h(15.73±1.31 ng/mL)、6h(18.66±1.57ng/mL)以及24h(16.19±1.08ng/mL)血清Irisin水平明显高于对照组血清Irisin水平(13.80±0.84ng/mL),(P0.05)。离心运动与对照组相比,运动后0h(16.37±1.34 ng/mL)、12h(25.37±1.63 ng/mL)、24h(15.60±1.63ng/mL)以及48h(34.45±3.30 ng/mL)血清Irisin水平明显高于对照组血清Irisin水平(13.80±0.84 ng/mL),(P0.01)。(4)运动后血清CTRP15血清水平的比较:向心运动组与对照组相比,运动后0h、6h、12h、24h、48h,血清CTRP15水平明显升高(P0.05)。离心运动除运动后6h,没有明显升高外,其它各时间点均有显著提高(P0.01)。结论:两种不同形式的运动(向心运动和离心运动)都能促进骨骼肌FNDC5的表达水平和血清Irisin、CTRP15的表达,向心运动后FNDC5与Irisin只出现了一次峰值,离心运动后FNDC5和Irisin出现两次峰值,且持续时间更长,释放的肌肉因子更多,效果更明显,更有利于运动减肥。
[Abstract]:Objective: exercise induced lipid metabolism in skeletal muscles to regulate muscle factors and thus promote fat oxidation is a hot topic in recent years. The aim of this study was to study the mechanism of exercise weight loss under the same oxygen intake. The effects of two muscle working forms (concentric exercise and centrifugal exercise) on serum Irisinine CTRP15 were studied in order to reveal the difference of lipid oxidation under the same energy metabolism level. Methods: 90 male Sprague-Dawley SD rats of 8 weeks old and weight of 200 卤20 g were used in this study. Rats were randomly divided into two groups: control group (n = 6), flat slope running group (n = 30) and downhill running group (n = 30). The exercise time of the exercise group was 60 minutes, and the slope of the downhill running group was-5 掳. Six rats in each group were collected blood samples from heart, gastrocnemius muscle and soleus muscle at 0 h, 6 h and 12 h after exercise for 24 h and 48 h, respectively. The FNDC5 level was measured at each time point. Serum Irisin and CTRP15 levels were measured by Elisa. The results of the study were as follows: (1) when the exercise time is the same and the running speed is different, There was no statistical difference in oxygen uptake between downhill and flat slope running rats (P 0.05) the expression of FNDC5 in gastrocnemius muscle increased significantly 6 hours after exercise in the concentric exercise group and the centrifugal exercise group, and the expression of FNDC5 in the gastrocnemius muscle in the concentric exercise group was significantly higher than that in the control group at 12 hours and 24 hours after the exercise, compared with that in the control group, and the expression of FNDC5 in the gastrocnemius muscle of the rats in the concentric exercise group was significantly higher than that in the control group. Compared with the control group, the serum Irisin level in the centrifuge exercise group was significantly higher than that in the control group, and the serum Irisin level in the concentric exercise group was significantly higher than that in the control group, and that in the concentric exercise group was higher than that in the control group. After exercise, the serum Irisin levels of 0h(15.73 卤1.31 ng / mL + 6 h, 18.66 卤1.57 ng / mL and 24h(16.19 卤1.08ng / mL) were significantly higher than those of the control group (13.80 卤0.84ng / mL). Compared with the control group, the serum Irisin levels of 0h(16.37 卤1.34ng / mL after exercise were significantly higher than those of the control group (13.80 卤0.84 ng / mL) after exercise: compared with that of the control group: the serum levels of CTRP15 in the concentric exercise group were significantly higher than those in the control group (25.37 卤1.63 ng / mL) and in the concentric exercise group and the control group, respectively, compared with those in the control group, and compared with those in the control group, the results showed that the serum Irisin level in the concentric exercise group was significantly higher than that in the control group (P < 0.05), and was significantly higher than that in the control group (P < 0.05). The serum levels of CTRP15 increased significantly at 0 h, 6 h, 12 h, 24 h and 48 h after exercise. Except for 6 hours after eccentric exercise, there was no significant increase in P0. 01, except that at other time points, there was a significant increase in P0. 01% at other time points. Conclusion: two different forms of exercise (concentric exercise and eccentric exercise) can promote the expression of FNDC5 in skeletal muscle and the expression of serum Irisinon CTRP15. There is only one peak value of FNDC5 and Irisin after concentric exercise, and two peaks of FNDC5 and Irisin after centrifugal exercise. And longer duration, the release of more muscle factors, more obvious effect, more conducive to exercise weight loss.
