均苯四甲酸降解菌PB-3的分离鉴定、降解特性及其降解途径研究

【摘要】:(1)均苯四甲酸是芳香族羧酸化合物中应用最为广泛的化合物之一,在化工合成及高分子行业中具有重要的作用。高浓度的均苯四甲酸废水因其可生化性差、COD值高、酸性强等特点,会给后续的污水处理带来很大困难,给自然环境造成很大危害。本研究从长青农药厂污水处理站的活性污泥中分离到了 一株能够高效降解均苯四甲酸的菌株将其命名为PB-3,该菌株能以均苯四甲酸为唯一碳源和能源进行生长。根据16S rRNA序列同源性分析,并结合菌株的表型特征和生理生化特性,将菌株PB-3初步鉴定为 Arthrobacter phenanthrenivorans。(2)菌株PB-3在LB培养基中生长良好。0~4h为菌株PB-3的延滞期,4~20h为菌株的对数期,菌株开始迅速生长,20 h以后开始进入稳定期。LB培养基中菌株的最适温度为30℃,最佳pH值为7.0。菌株为好氧菌,其生长量与装液量呈负相关。PB-3的最适盐浓度为10 g/L。在碳源利用试验中,菌株PB-3对葡萄糖和蔗糖的利用率较高,其次为果糖、甘露醇、木糖,几乎不能利用乳糖来进行生长;在氮源利用试验中,蛋白胨为氮源时,菌株PB-3长势最优,其次分别是氯化铵、硝酸钾、硫酸铵和尿素。在抗生素的耐受性试验中,菌株只对诺氟沙星具有抗性,对四环素、庆大霉素、卡那霉素、氯霉素、头孢曲松、青霉素、氧氟沙星、羧苄西林、氨苄西林、壮观霉素、链霉素均较为敏感。(3)菌株PB-3在无机盐培养基中的生长降解曲线表明,在64 h后PB-3能将400 mg/L的均苯四甲酸完全降解,并随着底物的降解,菌株的生长量逐步增加。在单因素试验中,PB-3降解均苯四甲酸的最适温度为30℃,最适pH为7.0。当均苯四甲酸的初始浓度从200 mg/L增加到1000 mg/L时,对菌株的降解能力没有明显的抑制作用。可以适当通过提高菌株PB-3的接种量来加快降解均苯四甲酸的速度。随着培养基中NaCl的浓度逐渐提高,对菌株降解均苯四甲酸能力的抑制越为强烈。以添加葡萄糖为外加碳源时,会延缓菌株对均苯四甲酸的降解。PB-3能够以1,2,4-苯三甲酸、邻苯二甲酸为唯一碳源并能够将其快速降解,但对间苯二甲酸的降解速度较为缓慢,且不能利用均苯三甲酸为碳源进行生长。PB-3对均苯四甲酸的降解符合Monod方程,其降解动力学方程为V=47.6S/(181 +S)。(4)通过LC-MS对菌株代谢均苯四甲酸的代谢中间体进行了分析,检测到了三种主要的代谢产物,分别为1,2,4-苯三甲酸、对苯二甲酸和2,5-二羟基对苯二甲酸。并对其代谢途径进行了初步推断。
[Abstract]:The main results are as follows: (1) phenyltetracarboxylic acid is one of the most widely used aromatic carboxylic acid compounds, which plays an important role in chemical synthesis and polymer industry. Because of its poor biodegradability, high COD value and strong acidity, high concentration of phenylenetetraformic acid wastewater will bring great difficulties to the subsequent sewage treatment and cause great harm to the natural environment. In this study, a strain named PB-3, was isolated from the activated sludge of sewage treatment station of Changqing Pesticide Plant, which can grow with phenyltetracarboxylic acid as the sole carbon source and energy source. According to the homology analysis of 16s rRNA sequence, combined with the phenotypic and physiological and biochemical characteristics of the strain, strain PB-3 was identified as Arthrobacter phenanthrenivorans.
