棉花pGUS-GhCOMT1基因融合载体构建及转化验证

【摘要】:为了深入研究GhCOMT1基因的功能,确定GhCOMT1基因对棉纤维细胞壁增厚的影响。本实验通过构建棉花木质素合成途径中的咖啡酸-O-甲基转移酶基因GhCOMT1的瞬时表达载体,用将pRTL2-GUS/NIa瞬时表达空载体连接GhCOMT1基因的目的片段,获得pGUS-GhCOMT1融合表达载体,并将其转化到棉花胚珠表皮细胞中进行表达。通过检测可以发现pGUS-GhCOMT1融合表达载体在洋葱表皮细胞及棉花胚珠表皮细胞中高效表达,表明棉花GhCOMT1基因对棉纤维的的伸长有显著的影响,同时为研究棉花纤维发育调控的分子机理提供理论基础。
[Abstract]:In order to study the function of GhCOMT1 gene, the effect of GhCOMT1 gene on cell wall thickening of cotton fiber was determined.In this experiment, the transient expression vector of caffeic acid-O-methyltransferase (GhCOMT1) gene in cotton lignin synthesis pathway was constructed, and the target fragment of GhCOMT1 gene was ligated with pRTL2-GUS/NIa transient expression empty vector. The fusion expression vector of pGUS-GhCOMT1 was obtained.It was transformed into cotton ovule epidermis cells for expression.It was found that pGUS-GhCOMT1 fusion expression vector was highly expressed in onion epidermis cells and cotton ovule epidermis cells, indicating that cotton GhCOMT1 gene had a significant effect on cotton fiber elongation.At the same time, it provides a theoretical basis for studying the molecular mechanism of cotton fiber development and regulation.
