茶树抗寒基因CsCBF1与CsICE1低温下的表达分析

【摘要】:为探明茶树抗寒转录因子基因CsCBF1与CsICE1的表达特性,以茶树幼苗为试验材料,根据其他已知植物的抗寒转录因子基因CBF1与ICE1序列,通过同源克隆法获得茶树中相对应的同源基因CsCBF1与CsICE1的保守区域,长度分别为456 bp与643 bp,经比对确定为CBF与ICE转录因子家族基因中CsCBF1与CsICE1型基因,因此命名为CsCBF1与CsICE1。荧光定量PCR分析CsCBF1与CsICE1基因在冷胁迫条件下的表达情况,发现其在4℃处理8 h时表达量最高;在茶树叶片中表达量显著高于根、茎、花。低温下CsCBF1与CsICE1基因表达显著增加,并且在抗寒性较强的种质(0306I和黄旦)中的表达显著高于抗寒性相对较弱的种质(0306F和0306D)。本研究结果为进一步探究CsCBF1与CsICE1基因在茶树耐低温胁迫过程中基因表达的调控机制奠定了基础。
[Abstract]:In order to investigate the expression characteristics of CsCBF1 and CsICE1 genes in tea plants, the chilling resistant transcription factor genes CBF1 and ICE1 sequences of other known plants were used as experimental materials. The conserved regions of the homologous genes CsCBF1 and CsICE1 in tea plants were obtained by homologous cloning. The length of CsCBF1 and CsICE1 were 456 BP and 643 BP, respectively, which were identified as CsCBF1 and CsICE1 type genes in CBF and ICE transcription factor family genes, so they were named CsCBF1 and CsICE1. The expression of CsCBF1 and CsICE1 genes under cold stress was analyzed by fluorescence quantitative PCR. It was found that CsCBF1 and CsICE1 gene expression was the highest at 4 鈩,
