从唾液中提取脱氧核糖核酸扩增血红蛋白β基因

【摘要】:目的:探索一种对人体无害、无痛、操作简便的获取基因组脱氧核糖核酸(deoxyribonucleic acid,DNA)的方法。方法:从唾液中提取基因组DNA,应用聚合酶链式反应(polymerase chain reaction,PCR)扩增血红蛋白β基因,采用含溴乙锭的琼脂糖凝胶电泳鉴定扩增产物。结果:从唾液中提取基因组DNA能够扩增出血红蛋白β基因。结论:唾液作为获取基因组脱氧核糖核酸的简易来源,取样方法简便、快捷,更适用于疾病研究中大规模群体筛查。
[Abstract]:Objective: to explore a simple and convenient method for obtaining genomic deoxyribonucleic acid (deoxyribonucleic acido DNA). Methods: genomic DNA was extracted from saliva. Hemoglobin 尾 gene was amplified by polymerase chain reaction (PCR) and agarose gel electrophoresis with ethidium bromide. Results: hemoglobin 尾 gene could be amplified by extracting genomic DNA from saliva. Conclusion: as a simple source of genomic DNA, saliva is a simple and rapid sampling method, which is more suitable for mass screening in disease research.
