BNC2基因多态性及其与鸡的银耳性状、产蛋性状的关系研究

【摘要】:BNC2基因在各种类型的细胞中广泛存在,可能参与RNA剪接过程。该基因与角质细胞增殖能力有关,在细胞终极分化中起重要作用。前人研究结果表明BNC2基因可能与鸡的色素合成相关,且参与了小鼠的卵巢发育。本论文将BNC2基因作为候选基因,分析了其多态性与鸡的银耳性状、产蛋性状之间的关系。主要研究结果如下:1.采用PCR方法,从鸡中获得了BNC2基因的三个转录本,长度分别为3020bp,1066bp,866bp,均有开放阅读框。2.对该BNC2基因进行蛋白的功能结构域分析,发现其结构域为Atrophin-1,并且其无跨膜区。对鸡与斑马鱼、青溕鱼、狗、大熊猫等动物BNC2基因进行了同源相似性分析,鸡与中国仓鼠、斑马鱼、青溕鱼属于同一分支,亲缘关系较近,与大熊猫、猴子、挪威鼠和家鼠亲缘关系较远。3.运用荧光定量PCR检测到BNC2基因在鸡的肝、肺、肾、下丘脑、卵巢、耳等12种组织中都有表达,其中在肺、下丘脑、卵巢组织的表达量较高,在肝、肾组织的表达量较低;在非银耳鸡中,耳的表达量更高,约是银耳鸡表达量的4倍。4.对清远麻鸡BNC2基因序列进行多态性位点筛查,共发现了20个SNPs。BNC2基因序列5’侧翼区12个多态位点与银耳性状和产蛋性状的关系,结果显示g.366AT,g.3411GA与鸡的银耳性状显著关联(P0.05);g.1600AG位点与鸡的产蛋性状关联显著(P0.05)。综上所述,通过PCR方法克隆了BNC2基因,获得了三个转录本,并且BNC2基因在鸡的肝、肺、肾、下丘脑、卵巢、耳等组织表达,其多态性可能与清远麻鸡的银耳性状、产蛋性状显著相关。
[Abstract]:BNC2 gene exists widely in various types of cells and may be involved in the RNA splicing process. The gene is related to the proliferation of keratinocytes and plays an important role in the ultimate differentiation of cells. Previous studies have suggested that BNC2 gene may be involved in the synthesis of chicken pigments and play an important role in the development of mouse ovaries. In this paper, BNC2 gene was used as a candidate gene to analyze the relationship between the polymorphism and the traits of Tremella and egg laying. The main results are as follows: 1. Using PCR method, three transcripts of BNC2 gene were obtained from chicken. The functional domain of the BNC2 gene was identified as Atrophin-1, and no transmembrane region. The homologous similarity analysis of BNC2 gene between chicken and zebrafish, green fish, dog, giant panda and other animals was carried out. Chicken and Chinese hamster, zebrafish, green fish belong to the same branch, close related to giant panda, monkey, etc. The relationship between Norway rat and house mouse is far away. 3. Fluorescence quantitative PCR was used to detect the expression of BNC2 gene in the liver, lung, kidney, hypothalamus, ovary, ear and other 12 tissues of chicken. The expression of BNC2 gene was higher in lung, hypothalamus and ovary, but lower in liver and kidney. In non-Tremella chickens, the expression level in the ear was 4. 4 times higher than that in the Tremella chicken. The polymorphic loci of BNC2 gene in Qingyuan hens were screened. A total of 12 polymorphic loci in 5 'flanking region of 20 SNPs.BNC2 gene sequences were found to be related to Tremella and egg laying traits. The results showed that g. 366AT. The correlation between g.3411GA and Tremella traits was significant (P0.05). There was a significant correlation between g.1600AG locus and egg laying traits (P0.05). To sum up, BNC2 gene was cloned by PCR, and three transcripts were obtained, and BNC2 gene was expressed in liver, lung, kidney, hypothalamus, ovary, ear and so on. Egg-laying traits were significantly correlated.
