杜仲Ref基因密码子偏好性分析

【摘要】:杜仲是世界上唯一的硬橡胶树种,是能够替代三叶橡胶的最具开发潜力的胶源植物。本研究以杜仲胶合成下游途径Ref基因为研究对象,运用Codon W和CUSPS等研究Ref基因的密码子组成特点及使用偏好性。研究结果表明,杜仲Ref基因的GC含量为49%,密码子第三位碱基的GC含量为59%;杜仲Ref基因有28个偏好使用的密码子,其中约60%(17个)的密码子以G/C结尾;与大肠杆菌、酵母、拟南芥和烟草的基因组密码子使用偏好性比较,杜仲与拟南芥基因组密码子的使用偏性更为接近,拟南芥更适合作为Ref转基因的受体。本研究为预测杜仲Ref基因的异源表达,适宜表达系统的选择以及Ref基因密码子的改造以提高其在宿主中的表达,进行有效的基因功能验证提供理论参考。
[Abstract]:Eucommia ulmoides is the only hard rubber tree in the world, and is the most developing potential gum source plant capable of replacing trifoliate rubber. In this study, the Ref gene of the downstream pathway of Eucommia ulmoides gum was used as the research object, and the Codon W and the CUSPS were used to study the codon composition and preference of the Ref gene. The results showed that the GC content of the Ref gene of the Eucommia ulmoides was 49%, the GC content of the third base of the codon was 59%, there were 28 codons in the Eucommia ulmoides's Ref gene, of which about 60% (17) of the codons were at the end of G/ C; and E. coli, the yeast, The using preference of the genomic codons of Arabidopsis and tobacco is closer to the use of the genomic codons of the eucommia and the Arabidopsis, and the arabidopsis is more suitable as a receptor for the Ref transgene. The study is to predict the heterologous expression of the Ref gene of the Eucommia ulmoides, the selection of the appropriate expression system and the transformation of the Ref gene codon to improve the expression of the Ref gene in the host, and to provide the theoretical reference for the validation of the gene function.
