结肠腺癌的发病机制探索及趋化因子CXC亚家族基因在结肠腺癌中的临床应用价值及机制研究

本研究分为四个部分:1、基于TCGA数据库的结肠腺癌差异基因及预后相关基因生物信息学分析;2、基于TCGA数据库的结肠癌患者趋化因子CXC亚家族基因表达的诊断和预后价值分析;3、广西医科大学一附院队列中CXCL8、CXCL3和CXCL1表达在结肠腺癌中的诊断与预后价值分析;4、利用小RNA干扰技术,分析SW620、RKO结肠癌细胞株中的CXCL8基因的生物学功能。第一部分 基于TCGA数据库的结肠腺癌差异基因及预后相关基因生物信息学分析目的:通过分析癌症基因组图谱(TCGA)数据库中结肠腺癌(COAD)的全基因组表达芯片数据集,鉴定COAD发生发展过程中的基因特征并评估其作为肿瘤生物标志物的潜力。材料与方法:从TCGA数据库下载COAD相关的mRNA测序数据集。使用R平台的edgeR和DESeq软件包筛选出COAD癌组织和癌旁正常结肠组织差异表达基因并用韦恩图过滤出交集,再用R平台的surviva1软件包校正影响因子后计算出与COAD预后相关的基因,用韦恩图过滤出与预后有关的差异基因,然后将这些不同作用的基因集合进行基因功能富集分析、信号通路富集分析、构建蛋白间和基因间相互作用网络等生...
