水稻SLG1和SLG2基因调控种子和器官大小的分子机理研究

水稻种子和叶片大小都是重要的农艺性状,它们的大小直接影响着水稻的产量。在植物中,器官大小通常是由细胞分裂和细胞扩展两个连续的生物学进程共同决定的。在单子叶模式植物水稻中,已经发现了许多调控水稻种子和叶片大小的因子,但这些器官大小因子的调控机制及相互之间的调控网络仍缺乏深入研究。实验室前期通过EMS诱变野生型水稻KYJ,分离获得突变体slgl-1,slgl-2和slg2,它们均具有种子窄,叶片窄的表型,过量表达 Slender Leaf and Grain 1(SLG1),Slender Leaf and Grain 2(SLG2)则产生增大的种子,表明SLG1和SLG2都是器官大小的正调节因子。本研究发现,SLG1和SLG2都通过降低细胞的分裂速率来调控种子和叶片的宽度。SLG1编码一种中介体复合物,SLG2编码一种组蛋白H3K4的去甲基化酶,蛋白都定位于细胞核中。SLG1和SLG2在穗子,种子及叶片中都有表达,在幼嫩组织中表达较强。遗传分析和蛋白互作结果表明,SLG1和SLG2作用于同一遗传途径调控种子和叶片宽度。烟草Co-IP,荧光素酶互补实验以及酵母双杂交实验结果显示,SLG1 ...
