Expression Pattern of Dmrt1Gene and the Effects of Octylphen

  本研究克隆了中国林蛙(Rana Chensinensis) Dmrtl (double-sex and mab-3related transcription factor1)基因,并检测了该基因在中国林蛙成体不同组织的表达以及在精巢中的定位。然后,将林蛙暴露于不同浓度的辛基酚(octylphenol, OP,10-8,10-7,10-6,10-5mol/L)后,检测精巢中Dmrtl mRNA的表达变化,探讨OP对Dmrt1基因表达的影响,分析Dmrt1可能的生理功能。采用反转录聚合酶链式反应(RT-PCR)和cDNA拼接技术,克隆中国林蛙Dmrt1基因,对其所编码的氨基酸序列信息,进化关系,蛋白质理化性质,二三级结构,亲水/疏水性,跨膜结构,信号肽,细胞定位进行分析。通过RT-PCR分析Dmrt1在成体精巢、卵巢、肝脏、肾脏和脑组织中的分布;对幼体性腺中肾复合体(GMC)中Dmrt1的表达变化也用RT-PCR进行分析;雌雄基因组中Dmrt1水平的差异用PCR技术进行简单评估。用荧光实时定量PCR检测不同OP浓度和不同暴露时间对Dmrt1mRNA表达的影响;用原位杂交技术检测Dmrt1m...
