【摘要】:目的 1、建立苯丙胺类为主的混合滥用药物成分定性定量分析方法。2、建立生物样品中AMP、MAMP及MDMA快速、灵敏检测方法。3、观察比对MDMA染毒家兔生命体征变化,并对急死与延缓死亡动物体内药物分布进行研究。 方法 1、薄层色谱扫描(TLCS)法:血浆样品经碱化,乙酸乙酯或环己烷萃取,薄层色谱扫描检测,Rf值结合光谱扫描图定性、外标两点法或标准曲线法定量分析。2、气相色谱-质谱联用(GC-MS)检测方法:体外检材乙醇溶解后GC-MS全扫描(full scan)定性定量分析。生物样品加入内标物SKF525A后碱化,乙醚萃取,TFA衍生化,GC-MS全扫描定性分析,选择离子扫描(SIM)定量分析。3、动物MDMA染毒与生命体征记录方法:家兔以10mg/kg剂量灌胃,BL-5生物机能实验系统记录从开始给药到动物被处死前的心电、血压和呼吸等主要生命体征变化。4、样品采集:动物分别于给药后2h和6h处死,迅速采集器官组织与体液检材,冷冻保存待检。 结果 1、11种滥用药物在DB-5色谱柱上能有效分离,,GC-MS检测线性关系良好。2、TLCS法对血浆中AMP与MDMA最低检出限为0.2 μg/斑点(S/N≥3),线性范围为0.2~15 μg/斑点。GC-MS法对血浆与肝脏中AMP、MAMP及MDMA检测线性范围分别为0.02μg/ml~2 .0 μg/ml和0.05μg/g~5 .0 μg/g,最低检出浓度分别为0.005 μg/ml和0.01 μg/g(S/N≥3)。3、家兔给药后,四肢出现间歇性震颤,血压升高,呼吸与心率加快,1h左右达到高峰,之后缓慢下降;给药2h后体内器官组织中药物浓度高于体液,其中以胃内容物、肺及脾含量最高,6h后组织与血液中药物浓度明显下降,而胆汁与尿液中药物浓度基本不变,胆汁中药物浓度高于其它组织。 结论 1、建立的混合滥用药物分析方法可用于11种组分快速、准确定性定量分析,为鉴别毒品、查证毒源及串并毒品案等禁毒、缉毒工作提供有力证据。2、TLCS与GC-MS分析方法分别适用于苯丙胺类中毒与滥用案例的快速鉴定。3、尿液为鉴定MDMA滥用的最佳检材;对滥用MDMA死亡者脏器组织与体液中药物含量测定,可对用药后急死或延缓死亡做出判断。
[Abstract]:Objective 1. To establish a qualitative and quantitative analysis method of amphetamine-based mixed drug abuse. 2. To establish a rapid and sensitive method for the detection of AMPMAMP and MDMA in biological samples, and to observe the changes of vital signs in rabbits exposed to MDMA. The distribution of drugs in animals with acute death and delayed death was studied. Methods 1. TLC scanning TLCSA: plasma samples were alkalized, extracted with ethyl acetate or cyclohexane, and their RF values were determined by TLC scanning and analyzed by spectral scanning. The external standard two-point method or standard curve method was used for quantitative analysis of .2. gas chromatography-mass spectrometry (GC-MS) was used to detect the quality and quantitative analysis of ethanol dissolved in vitro by GC-MS scanning. The biological samples were alkalized after adding the internal standard SKF525A, and then extracted by ether. The derivatization of TFAs was analyzed by GC-MS. The methods of animal MDMA exposure and vital signs recording were as follows: BL-5 biological function experiment system with 10mg/kg dose was used to record the electrocardiogram from the beginning of administration to the death of the animals. Changes of blood pressure and respiration. Sample collection: animals were killed at 2 h and 6 h after administration, organ tissue and body fluid samples were collected quickly and frozen to be preserved for examination.
