【摘要】:本论文以内蒙古正蓝旗青格勒图典型沙化梁地作为采样地,分梁底、梁中和梁顶3个样地采集羊柴(Hedysarum leave Maxim)根围土壤样品,通过Nested-PCR-DGGE和高通量测序技术,结合DGGE图谱分析、DNA克隆测序,研究AM真菌群落结构和物种多样性特征,为阐明沙化梁地AM真菌生态分布和功能提供依据。主要试验结果如下:1.DGGE研究结果表明,不同样地和土层,AM真菌具有不同DGGE指纹图谱特征,AM真菌丰度、优势度及其多样性指数均有差异,其中梁中和梁顶样地0-10 cm土层AM真菌丰度最高;多样性指数梁中样地0-10 cm土层最高。DGGE条带全部序列归为5属15种,即Glomus 10种、Rhizophagus 1种、Funneliformis 2种、Paraglomus 1种、Septoglomus1种;Glomus和Funneliformis是沙化梁地AM真菌优势属。聚类分析显示AM真菌群落具有丰富的物种多样性,相似性指数最高仅为0.54,说明梁地生境空间异质性对AM真菌物种多样性有显著影响。2.Illumina HiSeq 2500高通量测序结果表明,通过以97%相似度进行序列聚类,共得到486个OTUs,其中252个属于Glomeromycota,全部序列归于3纲,4目,10科和18个属。其中Glomeromycota在梁底的丰度高于梁顶,Glomeraceae在所有样品具有最高的相对丰富度。Sclerocytis和Glomus是所有样本中丰度最高的属,Clarodeoglomus,Rhizophagus和Gigaspora具有较高的相对丰度。Paraglomeraceae未在梁底发现,Geosiphonaceae未在梁中发现,Ambisporaceae和Pacisporaceae未在梁顶发现。3.冗余分析表明,土壤pH、有机碳(SOC)、全氮(TN)、总磷(TP)、有效氮(AVN)和有效磷(AVP)是影响AM真菌多样性和物种组成的重要影响因素。不同坡位和土层能够影响AM真菌群落组成和物种多样性,使其AM真菌分布具有空间异质性。
[Abstract]:This paper takes Inner Mongolia blue flag qinggelt typical Sandy Ridge as sampling points, the end of a beam, the collection of the Liang Zhonghe beam top 3 plots (Hedysarum leave Maxim) hedysarummaxim rhizosphere soil samples by Nested-PCR-DGGE and high-throughput sequencing technology, combined with the analysis of DGGE spectra, DNA cloning sequencing of AM fungal community structure and the characteristics of species diversity, provide the basis for clarifying the desertification ground AM fungi ecological distribution and function. The main results are as follows: the results of the 1.DGGE study, and equally soil fungi have AM fingerprint of different DGGE, AM fungi abundance, dominance and diversity index were different, in which the beam and beam top plot 0-10 cm AM the highest abundance of soil fungi; diversity index in 0-10 cm soil sample beam the highest.DGGE bands all sequences belonging to 5 genera and 15 species, namely Glomus 10, Rhizophagus 1, Funneliformis 2, Paraglomus 1 A kind of Septoglomus1, Glomus and Funneliformis are Sandy Ridge; AM fungi genera. Cluster analysis showed that the AM fungal community is rich in species diversity, the highest similarity index is only 0.54, that ridge habitat spatial heterogeneity has a significant impact on.2.Illumina HiSeq 2500 high-throughput sequencing results showed that the diversity of AM fungi species by sequence clustering with 97% similarity, there were 486 OTUs, 252 of which belong to Glomeromycota, all belong to sequence 3 classes, 4 orders, 10 families and 18 genera. Among them Glomeromycota in abundance is higher than that of top beam bottom beam, Glomeraceae has the highest relative abundance of.Sclerocytis and Glomus in all samples is abundance in all samples is the highest Clarodeoglomus, Rhizophagus and.Paraglomeraceae, the relative abundance of Gigaspora with high not found at the end of the beam, Geosiphonaceae was not found in the middle of the beam, Ambisporaceae and Pacis Poraceae is not found on the top of the beam.3. redundancy analysis showed that soil pH, organic carbon (SOC), total nitrogen (TN), total phosphorus (TP), available nitrogen (AVN) and phosphorus (AVP) is an important factor influencing the composition of AM fungal diversity and species. Different slopes and soil fungi can influence AM community composition and species diversity, the distribution of AM fungi with spatial heterogeneity.
