【摘要】:目的:本实验通过研究KLF9(Krüppel样转录因子9)在胰腺癌中的表达并研究其与胰腺癌患者临床病理参数及预后的关系,从而为胰腺癌的诊治提供新的研究思路。方法:本实验通过采集150例胰腺癌及其配对的癌旁组织标本,从中随机挑选28对标本运用实时荧光定量PCR(RT-PCR)技术,以了解胰腺癌及其配对的癌旁组织中的KLF9 m RNA的表达情况;随机挑选8对标本采用蛋白印迹技术(Western Blot)技术,并对所有的150对标本运用免疫组化技术检测胰腺癌及其配对癌旁组织中的KLF9蛋白的表达情况,并统计分析KLF9的表达水平与病人临床病理参数及生存率的关系。结果:实时荧光定量PCR结果显示,在28个配对的胰腺癌标本中,与癌旁正常胰腺组织相比,胰腺癌组织中KLF9 mRNA的表达水平明显下调(21/28)。蛋白免疫印迹结果显示,与癌旁正常对照胰腺组织相比,胰腺癌组织中KLF9蛋白表达水平明显下降。胰腺癌组织基因芯片(含150对胰腺癌组织和配对的正常的胰腺组织)免疫组化结果显示,KLF9在胰腺癌组织中的染色强度明显减弱。KLF9与病人临床病理参数分析显示,KLF9的表达水平与肿瘤的分化程度(p=0.008)和血管侵犯(p=0.006)显著相关。生存分析显示,KLF9阳性表达患者1、2、3年总生存率明显高KLF9阴性表达患者(41%、31%、16%vs.18%、6%、6%)、中位生存时间明显长于KLF9阴性表达患者(21.05个月vs.11.82个月)(均P0.05)。结论:通过RT-PCR、蛋白质免疫印迹、免疫组化技术显示KLF9在胰腺癌组织中低表达,其表达水平与肿瘤的分化程度及血管侵犯显著相关,同时KLF9与胰腺癌患者的生存时间也有相关性,由此可猜测KLF9可能介入了胰腺癌的发生发展过程。
[Abstract]:Objective: To investigate the effect of KLF9 (Kr ppel like transcription factor 9) expression in pancreatic carcinoma and to study its relationship with clinical pathological parameters and prognosis of patients with pancreatic cancer, so as to provide a new idea for the diagnosis and treatment of pancreatic cancer. Methods: This study collected 150 cases of pancreatic cancer and its adjacent tissue samples from 28 randomly selected specimens by real-time fluorescence quantitative PCR (RT-PCR) technology, the expression of KLF9 m RNA for pancreatic cancer and adjacent tissues; 8 randomly selected specimens using Western blot technique (Western Blot) technology, and the expression of all 150 samples on the use of immunohistochemical detection of pancreatic cancer and its adjacent tissues KLF9 protein, and statistical analysis of the expression of KLF9 and clinicopathological parameters and patient survival rate. Results: the results of real-time quantitative PCR showed that in 28 Pancreatic cancer specimens paired, compared with adjacent normal pancreatic tissue, the expression of KLF9 in pancreatic cancer tissues of mRNA was significantly reduced (21/28). Western blot results showed that compared with adjacent normal pancreatic tissue in the control, decreased level of expression of KLF9 protein in pancreatic cancer. Pancreatic cancer tissue microarray (including 150 in pancreatic cancer tissues and paired normal pancreatic tissue) immunohistochemistry showed that the staining intensity of KLF9 in pancreatic cancer patients significantly decreased.KLF9 and clinicopathological parameters analysis showed that KLF9 expression and tumor differentiation degree of (p=0.008) and vascular invasion (p=0.006). Survival analysis showed significant correlation, KLF9 the positive expression of 1,2,3 in patients with 5-year overall survival rate was significantly higher in patients with negative expression of KLF9 (41%, 31%, 16%vs.18%, 6%, 6%), the median survival time was significantly longer than that in patients with negative expression of KLF9 (21.05 vs.11.82 months) (P0. 05). Conclusion: by RT-PCR, Western blot, immunohistochemistry showed low expression of KLF9 in pancreatic cancer tissue, differentiation and vascular invasion of the tumor and its expression level was significantly related to patients with pancreatic cancer and KLF9 survival time is related to the speculation that KLF9 might be involved in the development and progression of pancreatic cancer process.
