【摘要】:目的:通过检测整合素亚单位α3(integrin subunitα3,ITGA3)在卵巢不同细胞系及组织中的表达水平,探讨ITGA3与卵巢上皮细胞癌间的关系,并分析ITGA3的表达与患者临床参数的相关性,为接下来研究其在卵巢癌发生、发展中的分子机制及靶向治疗提供实验依据。方法:(1)利用实时荧光定量PCR技术(q RT-PCR)检测ITGA3在正常卵巢上皮细胞系、各种卵巢癌细胞系、卵巢良性组织与卵巢上皮细胞癌组织中的m RNA表达水平。(2)免疫组化方法检测ITGA3在良性卵巢组织与上皮性卵巢癌组织中的蛋白表达情况。本研究用病理切片来自江苏大学附属医院、江苏大学附属人民医院及连云港市第二人民医院2013年至2016年术后病理存档的卵巢组织,共65例,其中上皮性卵巢癌组织38例,卵巢良性组织27例。收集患者临床资料,分析ITGA3的蛋白表达与卵巢癌患者临床参数的相关性。结果:(1)ITGA3在SKOV3和OVCAR-3两种卵巢癌细胞系中的m RNA表达水平高于正常卵巢上皮细胞HOSEPIC(P0.001)。(2)ITGA3在上皮性卵巢癌组织中的m RNA表达水平明显高于卵巢良性组织(P0.001)。(3)免疫组化结果显示:卵巢良性组织和上皮性卵巢癌组织中均有ITGA3蛋白的表达,但卵巢良性组中仅有4例呈阳性,而上皮性卵巢癌组中有26例阳性表达,两组间差异具有统计学意义(P=0.000)。在病理分期越高、分化越差的卵巢癌组织中ITGA3蛋白的高表达越明显(P=0.016,P=0.003)。结论:ITGA3有可能作为判断上皮性卵巢癌恶性发展程度的标志,为制定临床治疗方案提供参考依据,并有可能对患者的预后作出评估和成为卵巢癌治疗的靶点。
[Abstract]:Objective: to detect the expression of integrin subunit 伪 3ITGA3 in different ovarian cell lines and tissues. To investigate the relationship between ITGA3 and ovarian epithelial cell carcinoma, and to analyze the correlation between the expression of ITGA3 and the clinical parameters of the patients. Methods: using real-time fluorescence quantitative PCR technique to detect ITGA3 in normal ovarian epithelial cell line. Various ovarian cancer cell lines. Expression level of m RNA in benign ovarian tissue and ovarian epithelial cell carcinoma. Immunohistochemical method was used to detect the expression of ITGA3 in benign ovarian tissues and epithelial ovarian cancer tissues. From 2013 to 2016, 65 cases of ovarian tissues were recorded in the affiliated people's Hospital of Jiangsu University and the second people's Hospital of Lianyungang City, including 38 cases of epithelial ovarian cancer. The clinical data of 27 patients with benign ovarian tissue were collected. To analyze the correlation between the expression of ITGA3 protein and the clinical parameters of ovarian cancer patients. The expression of ITGA3 in SKOV3 and OVCAR-3 ovarian cancer cell lines was higher than that in normal ovarian epithelial cells. The expression of m RNA in epithelial ovarian carcinoma was significantly higher than that in benign ovarian carcinoma (P0.001. Immunohistochemical results showed that ITGA3 protein was expressed in benign ovarian tissues and epithelial ovarian cancer tissues. However, only 4 cases were positive in benign ovarian group and 26 cases in epithelial ovarian carcinoma group. The difference between the two groups was statistically significant. The higher the pathological stage was, the higher the pathological stage was. The higher the expression of ITGA3 protein in poorly differentiated ovarian cancer tissues, the more obvious the expression of ITGA3 protein was 0.016. Conclusion: TGA _ 3 may be used as a marker to judge the malignant development of epithelial ovarian cancer and provide reference for clinical treatment. It is possible to evaluate the prognosis of patients and become a target for ovarian cancer treatment.
