参芪补肺方对COPD大鼠KLF2、Nrf2及γ-GCS的影响

【摘要】:目的:观察参芪补肺方对COPD大鼠KLF2、Nrf2、y-GCS蛋白和mRNA的影响。方法:将健康、清洁级雌性Wistar大鼠40只,随机分为空白组、模型组、富露施组、中药组。采用香烟烟雾熏制和经鼻给予脂多糖法制作COPD大鼠模型。镜下观察大鼠肺组织病理形态学改变;RT-PCR方法检测COPD大鼠肺组织中KLF2、Nrf2、Y-GCSmRNA的表达;免疫组化方法检测COPD大鼠肺组织中KLF2、Nrf2、Y-GCS蛋白的表达。结果:1.HE染色后镜下观察大鼠肺组织,发现模型组、富露施组、中药组的肺组织改变与COPD病理改变相同。2.RT-PCR结果显示:模型组、富露施组、中药组大鼠肺组织KLF2、Nrf2、γ-GCS mRNA的表达高于空白组,经统计学处理有明显差异(P0.05);模型组大鼠肺组织KLF2、Nrf2、γ-GCSmRNA的表达明显高于富露施组、中药组,经统计学处理有意义(P0.05)。3.免疫组化结果显示:KLF2、Nrf2、Y-GCS蛋白见于肺泡、支气管、血管内皮细胞;模型组、富露施组、中药组肺组织中KLF2、Nrf2、γ-GCS蛋白的表达明显高于空白组,经统计学处理有明显差异(P0.05);富露施组、中药组KLF2、Nrf2、γ-GCS蛋白的表达低于模型组经统计学处理有差异(P0.05)。4.富露施组与中药组的肺组织中KLF2、Nrf2、Y-GCS mRNA和蛋白的表达相近,无统计学意义(P0.05)。结论:1.参芪补肺方可以减轻COPD大鼠肺组织的病理改变2.参芪补肺方可增加COPD大鼠肺组织中KLF2、Nrf2、γ-GCSmRNA和蛋白的表达,增加组织的抗氧化能力。
[Abstract]:Objective: To observe the effect of Shenqi tonifying lung Recipe on KLF2, Nrf2, y-GCS protein and mRNA in COPD rats. Methods: 40 healthy and clean female Wistar rats were randomly divided into blank group, model group, rich exposure group, Chinese medicine group. The rat model of COPD was made with cigarette smoke fumigating and transnasal lipopolysaccharide. The pathological morphology of lung tissue was observed under the microscope. The expression of KLF2, Nrf2 and Y-GCSmRNA in lung tissue of COPD rats was detected by RT-PCR method, and the expression of KLF2, Nrf2 and Y-GCS protein in lung tissue of COPD rats was detected by immunohistochemistry. Results: the lung tissue of rats was observed under the microscope after 1.HE staining, and the model group, rich exposure group, and the same.2.RT-PCR knot in the lung tissue of the Chinese medicine group and COPD pathological changes were found. The results showed that the expression of KLF2, Nrf2 and -GCS mRNA in the lung tissue of the rats in the model group and Fu Lu Shi group was higher than that in the blank group (P0.05). The expression of KLF2, Nrf2, and gamma -GCSmRNA in the lung tissue of the model group was significantly higher than that in the rich exposed group. The results of statistical processing showed significant (P0.05).3. immuno histochemical results showed: KLF2, Nrf2, Y-GCS protein was found in alveolar, bronchial, vascular endothelial cells, model group, rich exposure group, the expression of KLF2, Nrf2, gamma -GCS protein in the lung tissue of the Chinese medicine group was significantly higher than that in the blank group (P0.05). The expression of KLF2, Nrf2, and gamma -GCS protein in the group of Fu Lu Shi group was lower than that of the model group (P0.05). 4. rich dew KLF2, Shi Group and Chinese medicine group in the lung tissues of Nrf2, Y-GCS and mRNA protein expression are similar, no statistical significance (P0.05). Conclusion: 1. Shenqi Bufei recipe can reduce rat lung tissue pathological changes of COPD 2. Shenqi Bufei recipe can increase KLF2 and COPD of lung tissues in rats in Nrf2, the expression of gamma -GCSmRNA and protein, increase the antioxidant capacity of the tissues.
