高住高练对肥胖大鼠脂代谢的影响及机制研究

【摘要】:研究目的:从脂蛋白代谢、脂肪酸代谢(脂肪酸合成、转运、分解、氧化)、脂肪细胞因子与调节激素、PPARα调控等角度,系统研究高住高练对肥胖大鼠体重和脂代谢的影响及机制。 研究方法:建立肥胖大鼠高住高练0-4周模型,分为0周组、低氧安静1-4周组、低住低练1-4周组、高住高练1-4周组。水平动物跑台进行耐力训练,训练强度常氧25-26m/min,低氧20-21m/min,1h/d,6d/w,分别持续1-4周后取材测试。 主要结果:1)高住高练2-4周较低氧安静和低住低练显著降低肥胖大鼠的体重指数,2-4周降低大鼠体重和脂体比优于低氧安静;2)高住高练3、4周显著降低肥胖大鼠TC, TG,升高高密度脂蛋白,且4周效果优于低氧安静和低住低练;载脂蛋白AⅠ、AⅡ、脂蛋白酯酶升高,载脂蛋白B100、CⅢ降低,参与了脂蛋白代谢的调节;3)高住高练3周较低住低练显著降低肥胖大鼠肝脏脂肪酸合成酶基因表达。高住高练3周显著增加肥胖大鼠脂肪组织甘油三酯酶基因表达和激素敏感酯酶含量,且高于低住低练;高住高练4周提高肥胖大鼠腓肠肌肉碱脂酰转移酶、脂肪酸转位酶基因表达和脂肪LPL与腓肠肌LPL比值,且较低住低练更显著。高住高练2-4周降低肝脏脂肪酸结合蛋白和脂肪酸转运蛋白,且低于低住低练;4)高住高练3、4周较低住低练和低氧安静显著提高瘦素、瘦素受体、脂联素水平,降低内酯素水平;5)高住高练3、4周升高胰高血糖素、肾上腺素、生长激素水平,降低胰岛素和甲状腺素水平,且高住高练4周胰岛素显著低于低氧安静,甲状腺素显著低于低住低练,胰高血糖素显著高于低氧安静,高住高练3周生长激素高于低氧安静;6)高住高练组4周显著性升高大鼠腓肠肌PPARα mRNA水平;3周显著高于低住低练。 研究结论:高住高练3、4周较低住低练升高肥胖大鼠PPARα mRNA、瘦素和瘦素受体、脂联素、胰高血糖素、肾上腺素、生长激素,降低内脂素、胰岛素和甲状腺素,在其调节作用下,高住高练3、4周较低住低练改善脂蛋白代谢,抑制脂肪合成,促进脂肪分解和氧化,减少脂肪转运至胞内,导致脂代谢负平衡,从而减少体脂,降低体重指数。
[Abstract]:Objective: to study the effects and mechanisms of high altitude training on body weight and lipid metabolism in obese rats from the perspectives of lipoprotein metabolism, fatty acid metabolism (fatty acid synthesis, transport, decomposition, oxidation, adipocyte factor and regulating hormone PPAR 伪). Methods: the obese rats were divided into 0 week group, 1 4 weeks hypoxia quiet group, 1 4 weeks low living and low training group and 1 4 weeks high living high training group. Endurance training was performed on the horizontal animal running platform. The intensity of training was 25-26m / min, hypoxic 20-21m / min, 1h / d / min, 6d / w, respectively, and the samples were tested after 1-4 weeks.
