肠道菌群代谢产物对NAFLD的作用及其机制研究

目的:本研究旨在研究肠道菌群代谢产物在非酒精性脂肪性肝病(nonalcoholic fatty liver disease,NAFLD)发生发展中的作用及其作用靶点和分子机制,从而为NAFLD及其严重类型非酒精性脂肪性肝炎(nonalcoholic steatohepatitis,NASH)的防治提供新策略。方法:通过高脂肪高胆固醇饮食喂养大鼠/小鼠16周建立NASH动物模型,在第8周末对照组给予生理盐水灌胃处理,实验组分别给予肠道菌群代谢产物氧化三甲胺(trimethylamine N-oxide,TMAO)、吲哚丙酸(indole-3-propionic acid,IPA)及丁酸钠(sodium butyrate,NaB)灌胃干预。对肝脏组织切片进行HE染色,并通过病理评分评估其病变严重程度及实验组干预效果。收集粪便样品进行16S rDNA测序,分析肠道菌群多样性情况。检测血清内毒素、肝酶、脂质及脂蛋白的含量。采用免疫组织化学、免疫印迹及实时定量PCR(quantative real-time PCR,qPCR)等方法检测各组大鼠/小鼠肝脏组织脂质代谢、炎症以及应激损伤和纤维化等相...

1.3.3?TMAO对血清酶学改变的影响??我们检测了四组大鼠的血清肝酶水平。如图4所示,与Chow+Vehide组相??比,Chow+TMAO组大鼠血清酶学无显著改变;HFD+Vehicle组大鼠血清丙氨酸??氨基转移酶(alanine?transaminase,?ALT)、天冬氨酸氨基转移酶(aspartate??transaminase,?AST)及碱性磷酸酶(alkaline?phosphatase,ALP)水平均显著升高,??提示16周HFD饲喂可造成大鼠肝脏损伤。与HFD+Vehicle组相比,HFD+TMAO??组大鼠血清ALT及AST水平显著降低,ALP水平无显著改变。该结果表明TMAO??可减轻HFD诱导的大鼠肝损伤,与组织学结果一致。??
